Abstract Columbin, a key secondary metabolite in Tinospora sagittata, exhibits diverse pharmacological properties, yet its content shows notable variation across different varieties. This study investigated the molecular and biochemical basis behind such variation, analyzing columbin 32 T. sagittata varieties ranging from 5.40 mg/g to 46.09 mg/g. High low-columbin-content were selected for in-depth transcriptomic untargeted metabolomic analyses. Differential expression patterns of genes metabolites between leaves rhizomes these explored. Results revealed that involved biosynthesis pathway, namely, DXS, DXR, MCS, GPPS, PMK, exhibited significantly higher high At same time, HMGR HDR levels notably lower. Furthermore, columbin-content contained increased isoprene levels, crucial precursor. These findings suggest gene precursor variations likely drive differences insight into clarifies provides valuable direction breeding cultivating varieties, amplifying potential therapeutic applications.

Abstract This study used liquid chromatography-electrochemical detection (LC-ECD) combined with chromatography-tandem mass spectrometry (LC-MS/MS) to screen and characterize the antioxidant components of sweet tea ( Lithocarpus polystachyus ), evaluate quality samples from different origins. First, total phenolic flavonoid content tea, as well in vitro activity, including ABTS DPPH radical scavenging capacities ferric reducing ability, were determined. Samples origins demonstrated potent correlation coefficients exceeding 0.8 between activities. Then, fingerprints generated by LC-ECD 22 common peaks identified LC-MS/MS, most them being phloretin derivatives. The clustering heat map revealed differences various regions, major compounds trilobatin, phlorizin, 3-hydroxyphlorizin, its isomer. Furthermore, grey relational analysis confirmed a strong association (protocatechuic acid, epicatechin, isoquercitrin, trilobatin) trilobatin showing highest contribution, reaching 0.9. Finally, 11 quantitatively screened using providing guidance for control.
