Abstract Background To date, standardizing clinical predictive biomarkers for assessing the response to immunotherapy remains challenging due variations in personal genetic signatures, tumour microenvironment complexities and epigenetic onco‐mechanisms. Main Body Early monitoring of key non‐coding RNA (ncRNA) may help predicting efficacy cancer come up with standard ncRNA biomarkers. For instance, reduced miR‐125b‐5p level plasma non‐small cell lung patients treated anti‐PD‐1 predicts a positive outcome. The miR‐153 colorectal chimeric antigen receptor T lymphocyte (CAR‐T) therapy indicate activation T‐cell killing activity. miR‐148a‐3p miR‐375 levels forecast favourable responses CAR‐T‐cell B‐cell acute lymphoblastic leukaemia. In GPC3 peptide vaccine, serum miR‐1228‐5p, miR‐193a‐5p miR‐375‐3p were reported as good improved overall survival. Therefore, there is critical need further studies elaborate on that have potential predict early immunotherapy. Conclusions This review summarises important different immunotherapeutic modalities including monoclonal antibodies, small molecule inhibitors, vaccines CAR‐T cells. addition, concise discussion forthcoming perspectives provided, outlining technical approaches optimal utilisation immune‐modulatory tools therapeutic targets.

Among gynaecologic malignancies, ovarian cancer is one of the most dangerous, with a high fatality rate and relapse due to occurrence chemoresistance. Many researchers demonstrated that oxidative stress involved in tumour occurrence, growth development. Nuclear factor erythroid 2-related 2 (NRF2) an important transcription factor, playing role protecting against damage. Increased levels Reactive Oxygen Species (ROS) activate NRF2 signalling, inducing expression antioxidant enzymes, such as haem oxygenase (HO-1), catalase (CAT), glutathione peroxidase (GPx) superoxide dismutase (SOD), protect cells stress. However, activation responsible for development chemoresistance, inactivating drug-mediated normally leads cells’ death. In this review, we report evidence from literature describing effect on cancer, focus its function drug resistance, natural synthetic modulators protective normal preservation.

Most recurrences of lung cancer (LC) occur within 3 years after surgery, but the underlying mechanism remains unclear. Here, we collect LC tissues with shorter (<3 years, recurrence group) and longer (>3 non-recurrence recurrence-free survival. By using 16S sequencing, find that intratumor microbiome diversity is lower in group butyrate-producing bacteria are enriched group. The signature circulating DNA can accurately predict recurrence. We prove injection Roseburia promote subcutaneous tumor growth. Mechanistically, bacteria-derived butyrate promotes metastasis by increasing expression H19 cells through inhibiting HDAC2 H3K27 acetylation at promoter inducing M2 macrophage polarization. Depletion macrophages partially abolishes metastasis-promoting effect butyrate. Our results provide evidence for cross-talk between suggest potential prognostic therapeutic value microbiome.

Long noncoding RNAs (lncRNAs) are a group of with transcript lengths >200 nucleotides. Mounting evidence suggests that lncRNAs closely associated tumorigenesis. LncRNA H19 (H19) was the first lncRNA to function as an oncogene in many malignant tumors. Apart from established role promoting cell growth, proliferation, invasion, migration, epithelial-mesenchymal transition (EMT), and metastasis, it has been recently discovered also inhibits programmed death (PCD) cancer cells. In this review, we summarize mechanisms by which regulates PCD cells through various signaling pathways, molecular mechanisms, epigenetic modifications. Wnt/β-catenin pathway PI3K-Akt-mTOR pathway. It acts competitive endogenous RNA (ceRNA) regulation. The interaction between RNA-binding proteins (RBP) apoptosis cancer. Moreover, modifications, including DNA methylation histone involved H19-associated conclusion, via chemoresistance, highlighting promising research significance therapeutic target. We hope our study will contribute broader understanding development treatment.

Abstract Recently, extracellular vesicles (EVs) have been emphasized in regulating the hypoxic tumor microenvironment of breast cancer (BC), where tumor‐associated fibroblasts (TAFs) play a significant role. In this study, we describe possible molecular mechanisms behind pro‐tumoral effects EVs, secreted by hypoxia (HP)‐induced TAFs, on BC cell growth, metastasis, and chemoresistance. These are based long noncoding RNA H19 (H19) identified microarray analysis. We employed an silico approach to identify differentially expressed lncRNAs that were associated with BC. Subsequently, explored downstream regulatory mechanisms. isolated EVs from TAFs exposed HP, these denoted as HP‐TAF‐EVs henceforth. MTT, transwell, flow cytometry, TUNEL assays performed assess malignant phenotypes cells. A paclitaxel (TAX)‐resistant line was constructed, xenograft lung metastasis models established nude mice for vivo verification. Our observation revealed lncRNA significantly overexpressed, whereas miR‐497 notably downregulated HP induced activation stimulated secretion EVs. Coculture cells led increase TAX resistance latter. upregulated methylation delivering H19, which recruited DNMT1, thus lowering expression miR‐497. addition, H19‐containing hindered expression, enhancing tumorigenesis vivo. study presents evidence contribution reduction through recruitment turn promotes chemoresistance
