The gut microbiota can produce a variety of microbial-derived metabolites to influence tumor development. Tryptophan, an essential amino acid in the human body, be converted by microorganisms via indole pathway such as Indole-3-Lactic Acid (ILA), Indole-3-Propionic (IPA), Indole Acetic (IAA) and Indole-3-Aldehyde (IAld). Recent studies have shown that play key roles progression, they used adjuvant regimens for immunotherapy or chemotherapy. ABSTRACT The clinical application of doxorubicin (DOX) is limited due to its cardiotoxicity, which primarily attributed interaction with iron in mitochondria, leading lipid peroxidation and myocardial ferroptosis. This study aimed investigate the role gut microbiota‐derived metabolite, indole‐3‐lactic acid (ILA), mitigating DOX‐induced cardiotoxicity (DIC). Cardiac function, pathological changes, ferroptosis were assessed vivo. cardioprotective effects mechanisms ILA explored using multi‐omics approaches, including single‐nucleus RNA sequencing (snRNA‐seq) bulk RNA‐seq, further validated Nrf2 knockout mice. findings revealed that DOX treatment disrupted microbiota, significantly reducing levels tryptophan metabolite ILA. In DIC models, supplementation markedly improved cardiac reduced collagen deposition, mitigated atrophy. snRNA‐seq analyses indicated played a crucial Experimental data demonstrated decreased both mice DOX‐treated H9C2 cells, evidenced by restoration GPX4 SLC7A11 reduction ACSL4. Mechanistically, functions as ligand for aryl hydrocarbon receptor (AhR), upregulation expression. protective against abolished silencing AhR. Moreover, beneficial on eliminated Nrf2‐deficient conclusion, exerts therapeutic inhibiting through activation AhR/Nrf2 signalling pathway. Identifying microbial could offer viable strategies DIC.

This study investigates the protective effects of lactic acid, a metabolite Bifidobacterium, on non-alcoholic fatty liver disease (NAFLD) induced by high-sugar, high-fat diet (HFD) in mice, context gut-liver axis. A NAFLD mouse model was established using HFD, and different intervention groups were set up to Bifidobacterium its acid. The included control group, treatment Glyceraldehyde-3-P (G-3P) co-treatment NOD-like receptor family pyrin domain containing 3 (NLRP3) overexpression group. evaluation function lipid metabolism conducted liver-to-body weight ratio, histological staining, biochemical assays. Enzyme-linked immunosorbent assay (ELISA) performed measure inflammatory cytokines, western blotting used analyze expression NLRP3 inflammasome autophagy-related molecules. In vitro, an cell oleic with cells treated acid assess droplet accumulation inflammation. vivo findings indicated that, comparison CBX group (Control without antibiotic treatment), NAFLD/CBX (NAFLD administration) NAFLD/ABX exhibited increased higher accumulation, aggravated histopathological damage, elevated levels AST (Aspartate Aminotransferase), ALT (Alanine TC (Total Cholesterol), TG (Triglycerides), LDL-C (Low-Density Lipoprotein IL-6 (Interleukin-6), TNF-α (Tumor Necrosis Factor-alpha), IL-1β (Interleukin-1 beta), NLRP3-related molecules, while HDL-C (High-Density Cholesterol) significantly decreased. Intervention reversed these adverse changes. Further addition G-3P led more pronounced improvement symptoms, weakened Bifidobacterium. vitro results that Ole heightened inflammasome-related molecules relative Treatment effectively changes; however, effect overexpression. Abstract Background The Bifidobacterium genus is a prominent bacterial population in the gastrointestinal tract. Previous findings suggest that linked to tumor suppression mouse models of melanoma. Additionally, when combined with programmed death-ligand 1 (PD-L1) antibody, it can enhance anti-tumor treatment by increasing tumor-specific T-cell responses and promoting infiltration antigen-specific T cells into tumors. However, there lack studies on esophageal squamous cell carcinoma (ESCC). This study aimed investigate potential impact this cancer type. Methods We examined 213 samples from ESCC patients who underwent resection. presence was confirmed using quantitative polymerase chain reaction fluorescent situ hybridization (FISH). Patient overall survival (OS) analyzed positivity. Tumor-infiltrating lymphocytes (TILs) were evaluated via hematoxylin eosin stains, immunohistochemistry used assess death-1 (PD-1), PD-L1, cluster differentiation 8 (CD8), forkhead box P3 (FOXP3) expression. Nutritional status computed tomography scans. Results positivity showed no correlation patient OS or TIL levels; however, normal tissue associated lower FOXP3 levels, suggesting role upregulating immune responses. Patients present peritumor exhibited better skeletal muscle area volume. Conversely, poorer prognostic nutrition index values, likely due decreased albumin levels. Conclusion induce upregulated response more prevalent cases good nutritional status.
