Background Gut microbiota can significantly alter the risk or progression of cancer by maintaining gut immune system homeostasis. However, exact mechanism which and its metabolites influence colorectal tumorigenesis is unclear. Methods The roles tryptophan metabolite indole-3-acetic acid (IAA) in inflammation tumor development were investigated dextran sodium sulfate (DSS) azoxymethane (AOM)-DSS mouse models with without IAA supplementation Lactobacillus reuteri -produced IAA. Pregnane X receptor (PXR) knockout (KO) mice aryl hydrocarbon KO used to explore regulates interleukin (IL)-35 expression. IL-35 + cells stimulated vitro analyzed flow cytometry. Additionally, liquid chromatography-mass spectrometry. Results We found that IAA, a produced L. , inhibit colitis inducing expression immunosuppressant cells. HuREG3α IECtg had high levels intestinal microbiota-derived these resistant AOM-DSS-induced cancer. Patients also low peripheral blood Further studies revealed IAA-producing alleviated symptoms inhibited colon tumors macrophages, T cells, B produce IL-35. Finally, PXR completely abolished effects on Conclusion demonstrate improve precancerous inflammatory environment through IL-35, thereby inhibiting tumorigenesis, suggesting may be preventive factor for colitis-related cancers.

Purpose: We aimed to explore changes in plasma and urine indole lactic acid (ILA) levels the relationship between inflammation ILA chronic kidney disease (CKD) patients healthy people. Patients Methods: Forty-seven CKD 30 individuals were included this study. One-way ANOVA was used for variables with normal distribution homogeneous variance. A rank-sum test performed non-normally distributed variables. Correlation analyses using Pearson's or Spearman correlation analyses. Independent analyzed ordinal binary logistic regressions. Receiver operating characteristic (ROC) curve used. Results: Plasma positively correlated (r = 0.51, P < 0.01). BMI, age, creatinine, BUN, triglycerides, uric negatively hemoglobin levels. Urine albumin Ordered regression analysis showed that significantly (OR=4.49, 0.01), urinary (OR=2.14, urea (OR=1.43, 0.01) (OR=0.95, related. ROC curves indicated reliable predictors of CKD. plasma, (OR=5.92, 0.01; OR=2.79, Hs-CRP (OR=2.45, Conclusion: can potentially be as biomarkers inflammatory status. Background To investigate microbial characteristics across multibody sites from chronic pancreatitis (CP), through pancreatic benign tumors, to ductal adenocarcinoma (PDAC) at different stages. Methods 16S ribosomal RNA (rRNA) amplicon sequencing was conducted on saliva, duodenal fluid, and tissue obtained via endoscopic ultrasound-guided fine needle aspiration (EUS-FNA) of patients with CP, PDAC in stage I/II, III, IV. The neutral community model (NCM) assessed the assembly contribution MaAslin2 identified differential microbes. Results From CP IV patients, there a marked surge influence salivary microbiota constitution communities. Our observations revealed successive alteration species various bodily during tumorigenesis. Notably, Porphyromonas gingivalis , Treponema denticola Peptoanaerobacter stomatis Propionibacterium acidifaciens endodontalis Filifactor alocis etc., sequentially increased along progression tissue, whereas bacteria commonly used as probiotics Bifidobacterium breve Lactiplantibacillus plantarum declined. Furthermore, escalating trends tumorigenesis were mirrored fluid saliva. which intensified also found be enriched saliva short-term survival (STS) compared those long-term (LTS). Conclusions Salivary microorganisms prominent factors shaping landscape context. Further exploration these entities is imperative unravel their specific roles pathogenesis, potentially yielding insights for future therapeutic strategies.
