Pesticide toxicity is a global concern for honeybee populations, and understanding these effects at the molecular level critical. This study analyzed transcriptome of honeybees larval adult stages after chronic exposure to sublethal dose (0.0017 µg a.i./larva) spinetoram (SPI) during phase. Four groups were used: acetone-treated larvae (ATL), adults (ATAs), SPI-treated (STL), (STAs). In total, 5719 differentially expressed genes (DEGs) identified ATL vs. ATAs, 5754 STL STAs, 273 STL, 203 ATAs STAs (FC ≤ 1.5, p < 0.05). response SPI, 29 unique DEGs in 42 adults, with 23 overlapping between comparisons, suggesting linked SPI toxicity. Gene ontology analysis showed that affected metabolism-related lipid-transport-associated adults. KEGG pathway revealed an enrichment pathways predominantly associated metabolism, hormone biosynthesis, motor proteins STL. The transcriptomic data validated by qPCR. These findings demonstrated disrupts essential processes, potentially harming development behavior, underscoring need safer agricultural practices.

The adverse effects of plastic on adult animal and human health have been receiving increasing attention. However, its potential toxicity to fetuses has not fully elucidated. Herein, biodistribution polystyrene (PS) particles was determined after the maternal mice were orally given PS micro- and/or nano-particles with without surface modifications during gestational days 1 17. results showed that microplastics (MPs) nanoparticles (NPs) mainly emerged in alimentary tract, brain, uterus, placenta mice, only latter infiltrated into fetal thalamus. NPs carboxyl-modified induced differentially expressed genes enriched oxidative phosphorylation GABAergic synapse. Maternal administration gestation led anxiety-like behavior progenies their γ-aminobutyric acid (GABA) reduction prefrontal cortex amygdala at Week 8. N-Acetylcysteine (NAC), an antioxidant, alleviated particles-induced injury brain rescued progenies. Additionally, caused excessive ROS apoptosis neuronal cell lines, which prevented by glutathione supplementation. These suggested produced a negative effect inducing suppressing GABA synthesis brain. findings contribute estimating risk for health.

The insecticides spinosad and imidacloprid are neurotoxins with distinct modes of action. Both target nicotinic acetylcholine receptors (nAChRs), albeit different subunits. Spinosad is an allosteric modulator, that upon binding initiates endocytosis its target, nAChRα6. Imidacloprid triggers excessive neuronal ion influx. Despite these differences, low-dose effects converge downstream in the precipitation oxidative stress neurodegeneration.Using RNA-sequencing, we compared transcriptional signatures imidacloprid, at exposures. cause up-regulation glutathione S-transferase cytochrome P450 genes brain down-regulation fat body, whereas reduced expression immune-related observed both tissues. shows unique impacts on involved lysosomal function, protein folding, reproduction. Co-expression analyses revealed little to no correlation between affected by nAChRα6 expressing neurons, but a positive glial cell markers. We also detected experimentally confirmed body cells male germline cells. This led us uncover dysfunction following exposure, fitness cost spinosad-resistant (nAChRα6 null) males - testes, fertility.Spinosad share perturbations immunity-, energy homeostasis-, stress-related genes. Low doses other neurotoxic should be investigated for similar impacts. While target-site resistance mutation has evolved field, this may have cost. It is well known that modern agriculture would not be able to meet the current demand for food without help of pesticides. However, conventional pesticides have been proven extremely harmful environment, species they are applied to, and, ultimately, humans. As a result, bio-pesticides introduced in recent years and include natural substances control pests, such as biochemical pesticides, microorganisms used pest agents (microbial pesticides), pesticide produced by plants containing added genetic material, plant-incorporated protectants (PIPs). Although these products, their widespread use has led an increased presence raising concerns regarding potential impact on both environment human health. The aim our study was determine phyto- cytogenotoxicity caused two insecticides, certified ecological agriculture: one (BCP) other microbial (MP), which were three concentrations (the maximum recommended concentration manufacturers (MRFC), 1.5X MRFC, 2X MRFC) meristematic root tissues Allium cepa. results compared negative (tap water) positive (a chemical (CP) mainly Deltamethrin). Phytotoxic cytogenotoxic effects analyzed at time intervals (24 48 h) measuring length, growth percentage, inhibition percentage (phytotoxicity tests), micronuclei frequency chromosome aberrations (anaphase bridges, chromosomal fragments, anaphase delays, sticky chromosomes, laggard/vagrant chromosomes) (cytogenotoxicity analyses), respectively. tests conducted this showed insecticide provides greater safety when applied, even higher doses than those manufacturers, with insecticide, whose similar induced Deltamethrin. suggest insecticides clastogenic aneugenic effects, highlighting need prior testing any type before large-scale use, especially since A. cepa high sensitivity good correlation test systems, e.g., mammals.

Pupae were subjected SPI at LC20 LC50 concentrations, was evaluated using assessment, assays, transcriptome analysis utilizing ontology Kyoto Encyclopedia Genes Genomes (KEGG) enrichment. Transcriptome revealed 32 differentially expressed genes (DEGs) that common both vs. control (CK) CK comparisons, along with 24 DEGs unique comparison 76 comparison. KEGG pathway indicated substantial enrichment pathways associated drug metabolism, xenobiotic amino acid implying disruptions in detoxification mechanisms broader metabolic imbalances resulting from exposure. Morphological showed a normal structure group, while significant damage observed severe degeneration group. Detoxification assays activities cytochrome P450, glutathione S-transferase, carboxylesterase significantly increased (p < 0.05), indicating an initial induction responses; however, they declined LC50, suggesting inhibition or damage. The activity acetylcholinesterase markedly diminished more decline possible neurotoxicity. These findings indicate exposure causes alterations morphology, expression underscoring insecticide’s detrimental honey bee health.

Currently, insecticides that target nicotinic acetylcholine receptors (nAChR) are widely used. Studies on the sublethal effects of have found they can affect amount virus in insects. The mechanism by which insect load remain unclear. Here, we show nAChR targeting insecticide viral replication through immune deficiency (IMD) pathway. We demonstrate a low dose spinosad (6.8 ng/mL), acting as an antagonist to Drosophila melanogaster receptor α6 (Dα6), significantly elevates sigmavirus (DMelSV) titers adults melanogaster. Conversely, high (50 agonist Dα6, substantially decreases load. This bidirectional regulation levels is absent Dα6-knockout flies, signifying specificity spinosad's action Dα6. Furthermore, knockdown Dα6 results decreased expression genes IMD pathway, including dredd, imd, relish, and downstream antimicrobial peptide AttA AttB, indicating reduced innate response. Subsequent investigations reveal no significant difference between relish mutant flies Dα6-relish double mutants, suggesting pathway's role antiviral defense dependent Collectively, our findings shed light intricate interplay signaling pathway mediating immunity, highlighting potential for nAChR-targeting compounds inadvertently influence dynamics hosts. knowledge may inform development integrated pest management strategies consider broader ecological impact use.
