ABSTRACT Adrenoceptors (ARs) play a vital role in various physiological processes and are key therapeutic targets. The advent of optical control techniques, including optogenetics photopharmacology, offers the potential to modulate AR signaling with precise temporal spatial resolution. In this review, we summarize latest advancements signaling, encompassing optogenetics, photocaged compounds, photoswitchable compounds. We also discuss limitations current tools provide an outlook on next generation optogenetic photopharmacological tools. These emerging technologies not only enhance our understanding but pave way for developments.

Abstract Incorporation of non‐equilibrium actions in the sequence self‐assembly processes would be an effective means to establish bio‐like high functionality hierarchical assemblies. As a novel methodology beyond self‐assembly, nanoarchitectonics, which has as its aim fabrication functional materials systems from nanoscopic units through methodological fusion nanotechnology with other scientific disciplines including organic synthesis, supramolecular chemistry, microfabrication, and bio‐process, been applied this strategy. The application factors conventional is discussed on basis examples directed assembly, Langmuir–Blodgett layer‐by‐layer assembly. In particular, structures using bio‐active components such proteins or by combination bio‐components two‐dimensional nanomaterials, are described. Molecular photoswitches enable reversible external control of biological systems, nanomachines, and smart materials. Their development is driven by the need for low energy (green-red-NIR) light switching, to allow non-invasive operation with deep tissue penetration. The lack clear design principles adaptation optimization such systems limits further applications. Here we provide a rulebook tetra-ortho-chloroazobenzenes, an emerging class visible-light-responsive photochromes, elucidating role that substituents play in defining their key characteristics: absorption spectra, band overlap, photoswitching efficiencies, half-lives unstable cis isomers. We developed a new method for the conditional regulation of CRISPR/Cas9 activity in mammalian cells and zebrafish embryos using photochemically activated, caged guide RNAs (gRNAs). Caged gRNAs are generated by substituting four nucleobases evenly distributed throughout 5'-protospacer region with during synthesis. Caging confers complete suppression gRNA:dsDNA-target hybridization rapid restoration function upon optical activation. This tool offers simplicity programmability design, high spatiotemporal specificity embryos, excellent off-to-on switching, stability preserving ability to form Cas9:gRNA ribonucleoprotein complexes. novel tools control gene editing, thereby enabling investigation spatiotemporally complex physiological events obtaining better understanding dynamic regulation.

Abstract Precise control over signal amplification provides unparalleled opportunities for diverse applications. However, spatiotemporally controlled has not been realized because of the lack a design methodology. The aim this study was thus to develop conceptual approach remote at chosen time and site in living cells. This system constructed by re‐engineering functional units hybridization chain reaction (HCR) combination with upconversion photochemistry, resulting an activatable HCR high spatial temporal precision near‐infrared (NIR) light. As proof concept, we demonstrate spatially temporally resolved amplified imaging messenger RNA (mRNA) ultrahigh sensitivity vitro vivo. Furthermore, using targeting subcellular sites have developed new technique NIR‐initiated mRNA exclusively within specific organelle.
