Diketopyrrolopyrrole (DPP) is now widely used in the phototherapy of tumors, but most reported DPP molecules have no absorption near-infrared (NIR) region. Phototheranostics have emerged and flourished as a promising pattern for cancer theranostics owing to their precise photoinduced diagnosis therapeutic meet the demands of precision medicine. The information effect are directly determined by fluorescence imaging ability photothermal conversion efficiency (PCE) phototheranostic agents. Hence, how balance competitive radiative nonradiative processes agents is key factor evaluate effect. Herein, molecules named ICRs with high photostaibility rationally designed, exhibiting emission in second near-infrared window (NIR-II, 1000-1700 nm) PCE, which related strong donor-acceptor (D-A) interaction reorganization energy Noteworthily, ICR-Qu stronger D-A large-sized conjugated unit encapsulated nanoparticles exhibits PCE (81.1%). Stimulated emission depletion (STED) nanoscopy has broadened our horizons to unravel mysterious functions of cellular structures on an unparalleled nanometer scale. Nevertheless, intense laser power is a general prerequisite for STED super-resolution imaging with satisfactory resolution, inevitably leading severe photobleaching fluorophores, irreparable photodamage biosamples, and impaired quality. Herein, by modulating distinct acceptor units, series donor–acceptor–acceptor structured fluorescent acrylonitriles featured aggregation-induced (AIE) at low were systematically developed. These AIE luminogens (AIEgens) exhibited tunable near-infrared emissions (650–733 nm) high fluorescence quantum yields (QYs up 26.8%) in the solid state, significant Stokes shifts, large two-photon absorption cross-sections. They also typical solvatochromic effect ultrahigh QYs 98.4% low-polarity solvents. Furthermore, these lipophilic specifically lit lipid droplets (LDs) exceptionally photostability wash-free manner. By taking TPA-BT-ANBI as example, LDs excellent resolution 62 80 nm cytosolic nuclear LDs, respectively, saturation 0.83 MW/cm2 extended time-lapse LD dynamics was achieved. Subsequent use optical discrimination fatty liver tissues deep-tissue demonstrated. This study opens new avenues versatile photostable materials powers target-specific imaging.

Abstract To enhance the phototheranostic performance, agents with high reactive oxygen species (ROS) generation, good tumor‐targeting ability, and prolonged retention are urgently needed. However, symmetric donor–acceptor (D–A) type usually produce spherical nanoaggregates, leading to tumor targeting but inferior retention. Rod‐like nanoaggregates desired extend their in tumors; however, this remains a challenge. In particular, dynamically changeable shapes that integrate merits of different morphologies seldomly reported. Therefore, self‐assembled organic smart shape tunability designed here using an asymmetric D–A TIBT. The photoluminescence quantum yield solids is up 52.24% for TIBT also exhibits ROS generation corresponding (TIBT‐NCs). Moreover, dynamic self‐assembly changing from nanospheres nanorods occurrs TIBT‐NCs, contributing enhancement 0.55 0.72. addition, can be observed both vitro vivo, conferring TIBT‐NCs strong Finally, efficient photodynamic therapy inhibit growth achieved inhibition rate 90%. Abstract Photodynamic therapy (PDT) and photothermal (PTT) have emerged as promising treatment options, showcasing immense potential in addressing both oncologic nononcologic diseases. Single‐component organic phototherapeutic agents (SCOPAs) offer advantages compared to inorganic or multicomponent nanomedicine, including better biosafety, lower toxicity, simpler synthesis, enhanced reproducibility. Nonetheless, how further improve the therapeutic effectiveness of SCOPAs remains a challenging research area. This review delves deeply into strategies performance PDT PTT by optimizing structural design SCOPAs. These encompass augmenting reactive oxygen species (ROS) generation, mitigating dependence, elevating light absorption capacity, broadening region, enhancing conversion efficiency (PCE). The shortwave infrared (SWIR) region is an ideal spectral window for next-generation bioimaging to harness improved penetration and reduced phototoxicity. SWIR activity may also be accessed via supramolecular dye aggregation. Unfortunately, development of aggregation remains challenging. We propose a crystal-aided aggregate synthesis (CAASH) approach introduce layer rationality the J-aggregate successful water-soluble JV-aggregate with bisbenzannulated silicon rhodamine scaffold (ESi5). resulting SWIR-aggregates exhibit excellent stabilities toward organic solvents, pH, sonication, photobleaching, thiols, endogenous oxidative species. Notably, aggregates have high structure-dependent melting temperature ca. 330-335 K.
