Abstract Abnormal friction forces, compact matrix structure, and dynamic acidic microenvironment in osteoarthritis (OA) make it difficult to remodel cartilage regeneration by integrating prominent inflammatory responses anabolism. Here, pH‐responsive PSBMA‐ b ‐PColBP@KGN nanoparticles (PSPK NPs) are developed a simple supramolecular assembly process. PSPK NPs found reduce the coefficient of (0.028) under high loading pressure (10.41 MPa). The synergistic effect KGN release coupling deep penetration promotes generation microenvironment. Further, vivo studies confirm potential restoring homeostasis, as evidenced reduced osteophyte volume, aligned structures, abundant matrix. This study provides novel insights into effects release, penetration, hydration lubrication on OA treatment, offering strategy for design tissue‐adaptive promote tissue inflammation‐related diseases.

Development of nanomedicines that can collaboratively scavenge reactive oxygen species (ROS) and inhibit inflammatory cytokines, along with osteogenesis promotion, is essential for efficient osteoarthritis (OA) treatment. Herein, we report the design a ROS-responsive nanomedicine formulation based on fibronectin (FN)-coated polymer nanoparticles (NPs) loaded azabisdimethylphoaphonate-terminated phosphorus dendrimers (G4-TBP). The constructed G4-TBP NPs-FN size 268 nm are stable under physiological conditions, be specifically taken up by macrophages through FN-mediated targeting, dissociated in oxidative microenvironment. dendrimer having intrinsic anti-inflammatory property FN both antioxidative properties display integrated functions ROS scavenging, hypoxia attenuation, macrophage M2 polarization, thus protecting from apoptosis creating designed bone immune microenvironment stem cell osteogenic differentiation. Abstract Osteoarthritis (OA) is a prevalent, chronic degenerative disease that affects people worldwide. It characterized by the destruction of cartilage and inflammatory reactions. High levels reactive oxygen species (ROS) cause oxidative stress, which damages lipids, proteins, DNA, leading to cell damage death. Furthermore, ROS also induces production cytokines chemotaxis, further worsening response damaging resulted in limited movement. Herein, this work reports dual‐functional injectable hydrogel, can help inhibit inflammation scavenging provide lubrication reduce wear tear on joints. To create 3‐aminophenylboronic acid modified hyaluronic synthesized, then crosslinked with hydroxyl‐containing polyvinyl alcohol (PVA) construct dual dynamic covalent hydrogel oHA‐PBA‐PVA gel, Gel (HPP). The mentioned here possesses unique bond structure allows it be injected, self‐heal, lubrication. This innovative approach offers new possibility for treating osteoarthritis combining anti‐inflammatory effects.
