Flatworms (Platyhelminthes) are a basally branching phylum that harbours wealth of fascinating biology, including planarians with their astonishing regenerative abilities and the parasitic tape worms blood flukes exert massive impact on human health. PlanMine (http://planmine.mpi-cbg.de/) has mission objective providing both mineable sequence repository for also resource comparative analysis flatworm biology. While original release was entirely based transcriptomes, current transitions to more genomic perspective. Building recent availability high quality genome assembly planarian model species Schmidtea mediterranea, we provide gene prediction set now assign existing transcripts defined coordinates. The addition single cell bulk RNA-seq datasets greatly expands available expression information. Further, add transcriptomes from broad range other flatworms phylogeny-aware interface makes evolutionary comparisons accessible non-experts. At its core, continues utilize powerful InterMine framework consistent data annotations enable meaningful inter-species comparisons. Overall, 3.0 thus provides host new features biology wider research community.

ABSTRACT Planarians are a group of flatworms. Some planarian species have remarkable regenerative abilities, which involve abundant pluripotent adult stem cells. This makes these worms powerful model system for understanding the molecular and evolutionary underpinnings regeneration. By providing succinct overview taxonomy, anatomy, available tools orchestration regeneration, this Primer aims to showcase both unique assets questions that can be addressed with system.

Regenerative abilities vary dramatically across animals. Even amongst planarian flatworms, well-known for complete regeneration from tiny body fragments, some species have restricted while others are almost entirely incompetent. Here, we assemble a diverse live collection of 40 to probe the evolution head in group. Combining quantification species-specific head-regeneration with comprehensive transcriptome-based phylogeny reconstruction, show multiple independent transitions between robust whole-body and freshwater species. RNA-mediated genetic interference inhibition canonical Wnt signalling interference-sensitive bypassed all defects, suggesting that pathway is linked emergence defects. Our finding has roles reproductive system model Schmidtea mediterranea raises possibility trade-off egg-laying, asexual reproduction by fission/regeneration drives regenerative trait evolution. Although quantitative comparisons levels, yolk content strategy our remained inconclusive, they revealed divergent planarians. Altogether, study establishes planarians as taxon comparative research presents framework mechanistic abilities.

ABSTRACT Aquatic invertebrates play a pivotal role in (eco)toxicological assessments because they offer ethical, cost‐effective and repeatable testing options. Additionally, their significance the food chain ability to represent diverse aquatic ecosystems make them valuable subjects for studies. To ensure consistency comparability across studies, international (eco)toxicology guidelines have been used establish standardised methods protocols data collection, analysis interpretation. However, current primarily focus on limited number of invertebrate species, mainly from Arthropoda, Mollusca Annelida. These are suitable basic toxicity screening, effectively assessing immediate severe effects toxic substances organisms. For more comprehensive ecologically relevant assessments, particularly those addressing long‐term ecosystem‐wide impacts, we recommended use broader diversity since present choice taxa exacerbates scope ecotoxicological This review provides overview focusing major how assess impact chemicals environments. The work supports broad‐taxa approach environmental as it better represents natural populations inhabiting various ecosystems. Reproduction is a fundamental feature of life, it the way life persists across ages. This book offers new, wider vistas on this biological phenomenon, exploring how works through whole tree life. It explores facets such as asexual reproduction, parthenogenesis, sex determination and reproductive investment, with taxonomic coverage extended over all main groups - animals, plants including 'algae', fungi, protists bacteria. collates into one volume perspectives from varied disciplines zoology, botany, microbiology, genetics, cell biology, developmental evolutionary animal plant physiology, ethology integrating information common language. Crucially, aims to identify commonalties among phenomena, while demonstrating diversity even amongst closely related taxa. Its integrated approach makes valuable reference for students researchers, well an effective entry point deeper study specific topics.

Abstract To fully comprehend host-microorganism interactions, it is crucial to understand the composition and diversity of microbiome, as well factors that shape these characteristics. We investigated microbiome variation using freshwater planarian Schmidtea mediterranea , an invertebrate model in regeneration biology (eco-)toxicology, by exposing organisms various controlled conditions. The exhibited high variability, with most bacteria belonging Betaproteobacteria. Among diverse microbial communities, a few genera, such Curvibacter were consistently present, but significant alterations response changing relative abundance fluctuated during process, initially increasing before returning similar beginning situation. After applying external stress, other genera decreased. Variation over time, between different origin laboratories individuals, showed additional, yet to-be-identified, are present. Taking all results together, our study provides solid basis for future research focusing on bacterial functionality planarians invertebrates.

Abstract The planarian Schmidtea mediterranea can regenerate its entire body from small tissue fragments and is studied as regeneration model species. assembly functional analysis of genomes has proven challenging due high A/T content (70% A/T), repetitive nature, limited transferability routine laboratory protocols to their divergent biochemistry. Only few often fragmented genome assemblies are currently available, open challenges include the provision well-annotated chromosome-scale reference species other planarians for a comparative evolution perspective.
