Abstract Background Grip strength is a widely used and well-validated measure of overall health that increasingly understood to index risk for psychiatric illness neurodegeneration in older adults. However, existing work has not examined how grip relates comprehensive set mental outcomes, which can detect early signs cognitive decline. Furthermore, whether brain structure mediates associations between cognition remains unknown. Methods Based on cross-sectional longitudinal data from over 40,000 participants the UK Biobank, this study investigated behavioral neural correlates handgrip using linear mixed effect model mediation analysis. Results In analysis, we found greater was associated with better functioning, higher life satisfaction, subjective well-being, reduced depression anxiety symptoms while controlling numerous demographic, anthropometric, socioeconomic confounders. Further, females showed stronger most outcomes than males. baseline related performance at ~9 years follow-up, reverse much weaker. neuroticism, health, financial satisfaction were longitudinally subsequent strength. The results revealed widespread increased grey matter volume, especially subcortical regions temporal cortices. Moreover, volume these also correlated considerably mediated their relationship Conclusions Overall, largest population-scale neuroimaging dataset currently available, our findings provide well-powered characterization interplay strength, structure, may facilitate discovery possible interventions mitigate decline during aging.

Background Prior research has solely focused on the association between handgrip strength and risk of depression in single countries or general populations, but more knowledge is required from wider-spread cohorts target populations. Aims This study aimed to investigate using repeated measures adults aged 50 years over. Method Data were retrieved Survey Health, Ageing Retirement Europe (SHARE) waves 1, 2, 4, 5, 6 7, a hand dynamometer (Smedley, S Dynamometer, TTM) EURO-D 12-item scale, respectively. Time-varying exposure covariates modelled both Cox regression restricted cubic splines. Results A total 115 601 participants (mean age 64.3 (s.d. = 9.9), 54.3% women) followed-up for median 7.3 (interquartile range: 3.9–11.8) 792 459 person-years. During this period, 30 208 (26.1%) experienced depression. When as continuous variable, we observed an inverse significant each kg increase up 40 men 27 women. Conclusions Being physically strong may serve preventive factor older adults, limited maximum specific threshold

Genetic mutations that disrupt open reading frames and cause translation termination are frequent causes of human disease difficult to treat due protein truncation mRNA degradation by nonsense-mediated decay, leaving few options for traditional drug targeting. Splice-switching antisense oligonucleotides offer a potential therapeutic solution diseases caused disrupted inducing exon skipping correct the frame. We have recently reported on an exon-skipping oligonucleotide has effect in mouse model CLN3 Batten disease, fatal pediatric lysosomal storage disease. To validate this approach, we generated constitutively expresses Cln3 spliced isoform induced molecule. Behavioral pathological analyses these mice demonstrate less severe phenotype compared with model, providing evidence oligonucleotide-induced can efficacy treating This highlights how engineering through RNA splicing modulation be effective approach.

Alzheimer’s disease (AD), the most common cause of dementia, is a complex and multifactorial condition without cure at present. The latest treatments, based on anti-amyloid monoclonal antibodies, have only modest effect in reducing progression cognitive decline AD, whereas possibility preventing AD has become crucial area research. In fact, recent studies observed decrease dementia incidence developed regions such as US Europe. However, these trends not been mirrored non-Western countries (Japan or China), contributing factors this reduction remain unclear. Lancet Commission delineated constrained classification 12 risk across different life stages. Nevertheless, scientific literature pointed to over 200 factors—including sociodemographic, medical, psychological, sociocultural conditions—related development dementia/AD. This narrative review aims synthesize risk/protective Essentially, we found that vary between individuals populations, complicating creation unified prevention strategy. Moreover, dementia/AD explanatory mechanisms involve diverse array genetic environmental interact from early stages life. future, population-based cohorts are essential validate dementia. evidence would help develop public health policies

The relationship between handgrip strength (HGS) at baseline and subsequent depressive symptoms among middle-aged older Chinese adults remains highly uncertain. This research endeavored to investigate the effect of functional limitation on association these two variables. A total 5,684 participants aged ≥45 years from China Longitudinal Study Health Retirement (CHARLS) were enrolled, using data 2015 2018 waves. Functional was evaluated based participants' self-reported basic activities daily living (BADL) instrumental (IADL). Logistic regression models utilized analyze HGS symptoms, bootstrap analysis performed explore potential mediating role limitation. After adjusting for confounders, an inverse correlation detected (B = -0.885, p < 0.001), a positive found 1.054, 0.001). mediated size through a*b -0.933 (BCa 95% CI: -1.224, -0.642). had significant impact 18.9% overall association. Among senior population, accounted connection symptoms. Interventions targeting enhancement muscle should be regarded as crucial elements maintaining physical function preventing

Abstract Low handgrip strength, a hallmark measure of whole-body has been linked with greater odds cognitive decline and dementia; however, conflicting findings, which could be due to population characteristics choice tools, such for the assessment strength function domains, also exist. Therefore, we examined relationship comprehensive list tests assess domains using representative sample US older men women without neurodegenerative disorders as dementia. We analyzed cross-sectional data from National Health Nutrition Examination Survey (NHANES) between 2011 2014, study cohort 777 adults (380 397 women) above 60 years age. Handgrip was assessed dynamometer, while through Consortium Establish Registry Alzheimer’s Disease (CERAD) Word List Learning Test (WLLT), Recall (WLRT), Intrusion Count (WLLT-IC WLRT-IC), Animal Fluency (AFT), Digit Symbol Substitution (DSST). Sex-stratified multiple linear regression analyses were performed upon covariate adjustment age, ethnicity, socio-economic status, education, medical history, body mass index, physical activity, energy, protein, alcohol intake. Maximal positively associated scores, including CERAD WLLT ( P = 0.009, R 2 0.146) AFT 0.022, 0.024) in men, but not (CERAD WLLT: 0.253, AFT: 0.370). No significant associations WLLRT (men: 0.057, women: 0.976), WLLT-IC 0.671, 0.869), WLLRT-IC 0.111, 0.861), DSST 0.108, 0.091) observed. Dose–response curves exhibited prominent all after adjustment, no indication plateau these relationships. In conclusion, higher independently better learning ability novel verbal information fluency over age Longitudinal studies are required confirm whether muscle predicts changes sex-specific manner, this connection is affirmed possibility reverse causation declines activity levels preclinical phase
