Senescence impairs liver physiology, mitochondrial function and circadian regulation, resulting in systemic metabolic dysregulation. Given the limited research on effects of combined exercise an ageing liver, this study aimed to evaluate its impact metabolism, rhythms senescence-accelerated mouse-prone 8 (SAMP8) mouse-resistant 1 (SAMR1) mice. Histological, reverse transcription quantitative polymerase chain reaction (RT-qPCR) immunoblotting analyses were conducted, supplemented by transcriptomic data sets AML12 hepatocyte studies. Sedentary SAMP8 mice exhibited decreased muscle strength, reduced complex I levels increased lipid droplet accumulation. In contrast, mitigated strength loss, upregulated proteins involved complexes (CIII, CIV, CV) Bmal1 messenger RNA (mRNA) expression liver. These molecular adaptations are associated with healthier phenotypes may influence cellular longevity. Notably, elevated content aged was post-exercise, indicating benefits even after a relatively short intervention. The regimen did not improve aerobic capacity, likely due low volume brief duration running. Moreover, no significant observed SAMR1 mice, possibly because training intensity insufficient for younger, animals. findings underscore potential endurance attenuate age-related dysfunction, particularly populations.

Abstract Peroxisome proliferator-activated receptor gamma coactivator-1 (PGC-1) family (PGC-1s), consisting of three members encompassing PGC-1α, PGC-1β, and PGC-1-related coactivator (PRC), was discovered more than a quarter-century ago. PGC-1s are essential coordinators many vital cellular events, including mitochondrial functions, oxidative stress, endoplasmic reticulum homeostasis, inflammation. Accumulating evidence has shown that implicated in diseases, such as cancers, cardiac diseases cardiovascular neurological disorders, kidney motor system metabolic disorders. Examining the upstream modulators co-activated partners identifying critical biological events modulated by downstream effectors contribute to presentation elaborate network PGC-1s. Furthermore, discussing correlation between well summarizing therapy targeting helps make individualized precise intervention methods. In this review, we summarize basic knowledge regarding molecular regulatory network, discuss physio-pathological roles human review application PGC-1s, diagnostic prognostic value several therapies pre-clinical studies, suggest directions for future investigations. This presents immense potential treatment hopefully facilitates promotion new therapeutic targets.

Abstract Physical activity, including walking, has numerous health benefits in older adults, supported by a plethora of observational and interventional studies. Walking decreases the risk or severity various outcomes such as cardiovascular cerebrovascular diseases, type 2 diabetes mellitus, cognitive impairment dementia, while also improving mental well-being, sleep, longevity. Dose-response relationships for walking duration intensity are established adverse outcomes. Walking’s favorable effects on factors attributed to its impact circulatory, cardiopulmonary, immune function. Meeting current physical activity guidelines briskly 30 min per day 5 days can reduce several age-associated diseases. Additionally, low-intensity exercise, exerts anti-aging helps prevent age-related making it powerful tool promoting healthy aging. This is exemplified lifestyles individuals Blue Zones, regions world with highest concentration centenarians. other activities contribute significantly longevity these regions, being an integral part their daily lives. Thus, incorporating into routines encouraging walking-based interventions be effective strategy aging all populations. The goal this review provide overview vast consistent evidence supporting specific focus discuss outcomes, prevention Furthermore, will delve molecular cellular mechanisms aging, providing insights underlying biological through which beneficial effects.

Mitochondria plays an essential role in regulating cellular metabolic homeostasis, proliferation/differentiation, and cell death. Mitochondrial dysfunction is implicated many age-related pathologies. Evidence supports that the of mitochondria decline mitochondrial DNA copy number negatively affect ovarian aging. However, mechanism aging still unclear. Treatment methods, including antioxidant applications, transplantation, emerging biomaterials, advanced technologies, are being used to improve function restore oocyte quality. This article reviews key evidence research updates on damage pathogenesis aging, emphasizing may accelerate lead senescence as well exploring potential methods for using mechanisms slow down

Abstract Cardiorespiratory fitness (CRF) not only reflects an individual’s capacity to perform physical activities but also encapsulates broader effects on the basic biology of aging. This review aims summarize evidence influence CRF overall and site-specific cancer risks. It delves into biological mechanisms through which may exert its effects, explores clinical implications these findings, identifies gaps in current base, suggests directions for future research. The synthesis findings reveals that higher levels (general threshold > 7 METs) are consistently associated with a reduced risk range cancers, including head neck, lung, breast, gastrointestinal, particularly pancreatic colorectal, bladder, incidence mortality, potentially stomach liver, bile duct, gall bladder cancers. These inverse associations between do generally differ across age groups, sex, race, or adiposity, suggesting universal protective effect CRF. Nonetheless, linking skin, mouth pharynx, kidney, endometrial cancers is limited inconclusive. Conversely, be linked increased prostate hematological malignancies, such as leukemia myeloma, although still conclusive. appears play significant role reducing several various mechanisms, inflammation reduction, immune system enhancement, hormonal regulation, metabolic improvements. Overall, enhancing regular activity offers vital, accessible strategy extending health span. Future research should aim fill existing regarding specific elucidate detailed dose–response relationships risk. Studies needed causal mechanistic pathways outcomes.
