Functional hydrogels are used for numerous biomedical applications such as tissue engineering, wound dressings, lubricants, contact lenses and advanced drug delivery systems. Surgical resection remains the prefer option for bladder cancer treatment. However, effectiveness of surgery is usually limited high recurrence rate and poor prognosis. Consequently, intravesical chemotherapy synergize with immunotherapy in situ an attractive way to improve therapeutic effect. Herein, a combined strategy based on thermo-sensitive PLEL hydrogel drug delivery system was developed. GEM loaded instilled kill tumor cells directly, then incorporated CpG injected into both groins subcutaneously promote immune responses GEM. The results demonstrated that had sol-gel phase transition behavior response physiological temperature presented sustained release, PLEL-assisted combination therapy could have better suppression effect stronger immunostimulating vivo. The severe side effects of systemic chemotherapy for cervical cancer encourage the use topical intravaginal drug delivery systems. 5-fluorouracil, 5-FU, is an anticancer accepted in clinical therapy colorectal, gastric, and hepatocellular carcinoma. Abstract Patients with colorectal cancer (CRC) and diffuse peritoneal metastasis (PM) are not eligible for surgical intervention. Thus, palliative treatment remains the standard of care in clinical practice. Systemic chemotherapy fails to cause drug accumulation at lesion sites, while intraperitoneal (IPC) is limited by high clearance rates associated complications. Given poor prognosis, a customized OxP/R848@PLEL hydrogel delivery system has been devised improve benefit advanced CRC PM. This distinguished its simplicity, security, efficiency. Specifically, PLEL exhibits excellent injectability thermosensitivity, enabling formation depots within abdominal cavity, rendering it an optimal carrier IPC. Oxaliplatin (OxP), first‐line CRC, cytotoxic enhances immunogenicity tumors inducing immunogenic cell death.
