The skin, including the hypodermis, is largest body organ and in constant contact with environment. Neurogenic inflammation result of activity nerve endings mediators (neuropeptides secreted by development inflammatory reaction skin), as well interactions other cells such keratinocytes, Langerhans cells, endothelial mast cells. activation TRPV–ion channels results an increase calcitonin gene-related peptide (CGRP) substance P, induces release pro-inflammatory contributes to maintenance cutaneous neurogenic (CNI) diseases psoriasis, atopic dermatitis, prurigo rosacea. Immune present skin (mononuclear dendritic cells) also express TRPV1, their directly affects function. TRPV1 mediates communication between sensory immune increasing (cytokines neuropeptides). Understanding molecular mechanisms underlying generation, modulation neuropeptide neurotransmitter receptors can aid effective treatments for disorders.

Abstract Atopic dermatitis (AD) is a chronic skin disease caused by immune dyshomeostasis and accompanied severe pruritus. Although oxidative stress mechanical scratching can aggravate AD inflammation, treatment targeting often overlooked, the efficiency of mechano-chemically synergistic therapy remains unclear. Here, we find that enhanced phosphorylation focal adhesion kinase (FAK) associated with scratch-exacerbated AD. We then develop multifunctional hydrogel dressing integrates modulation FAK inhibition to synergistically treat show adhesive, self-healing antimicrobial suitable for unique bacterial environment skin. demonstrate it scavenge intracellular reactive oxygen species reduce mechanically induced intercellular junction deficiency inflammation. Furthermore, in mouse models controlled scratching, alleviates symptoms, rebuilds barrier, inhibits These results suggest integrating scavenging could serve as promising treatment.

Atopic dermatitis (AD) is a chronic inflammatory skin disease, accompanied by itching and swelling. The main pathological mechanism of AD related to the imbalance between Type 2 helper cells (Th2 cells) 1 (Th1 cells). Currently, no safe effective means treat prevent are available; moreover, some treatments have side effects. Probiotics, such as strains Lactobacillus , can address these concerns via various pathways: i) facilitating high patient compliance; ii) regulating Th1/Th2 balance, increasing IL-10 secretion, reducing cytokines; iii) accelerating maturation immune system, maintaining intestinal homeostasis, improving gut microbiota; iv) symptoms AD. This review describes treatment prevention using 13 species . commonly observed in children. Therefore, includes higher proportion studies on children fewer adolescents adults. However, there also that do not improve even worsen allergies In addition, subset genus relieve has been identified vitro future should include more vivo randomized controlled clinical trials. Given advantages disadvantages mentioned above, further research this area urgently required.

Psychological stress is a crucial factor in the development of many skin diseases, and stigma caused by disorders may further increase psychological burden, forming vicious cycle leading to diseases. Therefore, understanding relationship between diseases necessary. The skin, as vital interface with external environment, possesses its own complex immune system, neuroendocrine system plays central role response body. Stress-induced alterations can also disrupt delicate balance cells inflammatory mediators dysregulation increased susceptibility various Stress affect barrier function, impair wound healing, promote release pro-inflammatory cytokines, thereby exacerbating existing such psoriasis, atopic dermatitis, acne, urticaria. In present review, we explored intricate from neuroendocrine-immune interaction perspective. We occurrence context stress, models for impact on function relevant epidemiological studies clinical trials. Understanding perspective provides comprehensive framework targeted interventions new insights into diagnosis treatment

Atopic dermatitis (AD) is a chronic, relapsing immunoinflammatory skin condition characterized by sensations such as pruritis, pain, and neuronal hypersensitivity. The mechanisms underlying these are multifactorial involve complex crosstalk among several cutaneous components. This review explores the role components play in pathophysiology of atopic dermatitis. In intercellular spaces, sensory nerves interact with keratinocytes immune cells via myriad mediators receptors. These interactions generate action potentials that transmit pruritis pain signals from peripheral nervous system to brain. Keratinocytes, most abundant cell type epidermis, key effector cells, triggering neurons elicit inflammation. Filaggrin expression reduced dermatitis, causing weakened barrier elevated pH. Fibroblasts main dermis and, appear reduce keratinocyte differentiation, further weakening barrier. mast promote inflammation while dermal dendritic attenuate Inflammatory cytokines chemokines major AD pathogenesis. Similar to chronic pain, itch is frequently linked neural sensitization, a phenomenon wherein the nervous system becomes hypersensitive stimuli. This process of sensitization orchestrated by various signaling pathways and mediators in both peripheral central systems. At level system, inflammation neuroimmune interactions induce plastic changes nerve fibers, thereby amplifying transmission signaling. Neural occurs at spinal cord brain levels. cord, it involves hyperactivity itch-activating pathways, dysfunction inhibitory circuits, attenuation descending supraspinal pathways. In brain, manifests as structural functional itch-associated areas networks. Currently, we have diverse array neuroimmune-modulating therapies targeting mechanisms help with providing relief patients itch. Itch research dynamic continually evolving field, grow our understanding mechanisms, so will therapeutic toolbox. Further studies exploring context are needed.
