Pterostilbene (PST) is a naturally derived stilbene compound in grapes, blueberries, and other fruits. It also natural dietary with wide range of biological activities such as antioxidant, anti-inflammatory, antitumor, so on. Structural modifications based on the chemical scaffold pterostilbene skeleton are great importance for drug discovery. In this study, skeletons were used to design novel anti-inflammatory compounds high activity low toxicity. A total 30 new found synthesised, their safety screened. Among them, E2 was most active (against NO: IC50 = 0.7 μM) than celecoxib. Further studies showed that exerted by blocking LPS-induced NF-κB/MAPK signalling pathway activation. vivo experiments revealed had good alleviating effect acute colitis mice. conclusion, may be promising lead compound.

Abstract The ageing process is a systemic decline from cellular dysfunction to organ degeneration, with more predisposition deteriorated disorders. Rejuvenation refers giving aged cells or organisms youthful characteristics through various techniques, such as reprogramming and epigenetic regulation. great leaps in rejuvenation prove that not one-way street, many rejuvenative interventions have emerged delay even reverse the process. Defining mechanism by which roadblocks signaling inputs influence complex programs essential for understanding developing strategies. Here, we discuss intrinsic extrinsic factors counteract cell rejuvenation, targeted core mechanisms involved this Then, critically summarize latest advances state-of-art strategies of rejuvenation. Various methods also provide insights treating specific ageing-related diseases, including reprogramming, removal senescence (SCs) suppression senescence-associated secretory phenotype (SASP), metabolic manipulation, stem cells-associated therapy, dietary restriction, immune heterochronic transplantation, etc. potential applications therapy extend cancer treatment. Finally, analyze detail therapeutic opportunities challenges technology. Deciphering will further into anti-ageing disease treatment clinical settings.

Abstract Diet, serving as a vital source of nutrients, exerts profound influence on human health and disease progression. Recently, dietary interventions have emerged promising adjunctive treatment strategies not only for cancer but also neurodegenerative diseases, autoimmune cardiovascular metabolic disorders. These demonstrated substantial potential in modulating metabolism, trajectory, therapeutic responses. Metabolic reprogramming is hallmark malignant progression, deeper understanding this phenomenon tumors its effects immune regulation significant challenge that impedes eradication. Dietary intake, key environmental factor, can tumor metabolism. Emerging evidence indicates might affect the nutrient availability tumors, thereby increasing efficacy treatments. However, intricate interplay between pathogenesis other diseases complex. Despite encouraging results, mechanisms underlying diet-based remain largely unexplored, often resulting underutilization management. In review, we aim to illuminate various interventions, including calorie restriction, fasting-mimicking diet, ketogenic protein restriction high-salt high-fat high-fiber aforementioned diseases. We explore multifaceted impacts these encompassing their immunomodulatory effects, biological impacts, molecular mechanisms. This review offers valuable insights into application therapies

Cyclic GMP-AMP synthase (cGAS) is the major sensor for cytosolic DNA and activates type I interferon signaling plays an essential role in antitumor immunity. However, it remains unclear whether cGAS-mediated activity affected by nutrient status. Here, our study reports that methionine deprivation enhances cGAS blocking its methylation, which catalyzed methyltransferase SUV39H1. We further show methylation chromatin sequestration of a UHRF1-dependent manner. Blocking immunity suppresses colorectal tumorigenesis. Clinically, human cancers correlates with poor prognosis. Thus, results indicate stress promotes activation via reversible suggest potential therapeutic strategy targeting cancer treatment.

An increasing number of studies suggest that diet plays an important role in regulating aging processes and modulates the development most age-related diseases. The aim this review is to provide overview relationship between nutrition critical age-associated diseases.A literature was conducted survey recent pre-clinical clinical findings related nutritional factors modulation fundamental cellular molecular mechanisms their prevention genesis diseases aging.Studies show cardiovascular cerebrovascular diseases, neurodegenerative cognitive impairment dementia can be slowed down or prevented by certain diets with anti-aging action. The protective effects diets, at least part, may mediated beneficial macro- (protein, fat, carbohydrate) micronutrient (vitamins, minerals) composition.Certain such as Mediterranean diet, play a significant healthy preventing onset improving process itself. This latter strengthened incorporating fasting elements into diet. As dietary recommendations change age, should taken consideration well, when developing tailored needs elderly individuals. Future ongoing on complex interventions translating results preclinical investigations are expected lead novel guidelines for older adults near future.

Diet may influence biological aging and the discrepancy (∆age) between a subject's age (BA) chronological (CA). We aimed to investigate correlation of dietary flavonoids with ∆age organs (heart, kidney, liver) whole body.A total 3193 United States adults were extracted from National Health Nutrition Examination Survey (NHANES) in 2007-2008 2017-2018. Dietary intake was assessed using 24-h recall method. Multiple linear regression analysis performed evaluate association body. BA computed based on circulating biomarkers, resulting tested as an outcome analysis. The body, heart, liver inversely associated higher (the body β = - 0.58, cardiovascular 0.96, 3.19) after adjustment for variables. However, positively related renal (β 0.40) participants chronic kidney disease (CKD). Associations influenced by population characteristics, such age, health behavior, or diseases. Anthocyanidins, isoflavones flavones had strongest inverse associations among all subclasses.Flavonoids contributes delaying process, especially organ, which be beneficial reducing long-term risk disease.
