Arecoline is an alkaloid extracted from betel nut, which has various pharmacological effects. In the present study, we showed that arecoline aggravated experimental acute ulcerative colitis (UC) induced by dextran sodium sulfate (DSS) in mice. We measured body weight and colon length, evaluated disease activity index, pathology sections, levels of colonic inflammatory factors. exacerbated clinical signs UC response The results 16S rRNA sequencing fecal samples a significant decrease percentage probiotic bacteria Ligilactobacillus, Limosilactobacillus Lactobacillus increase conditionally pathogenic Odoribacter Bacteroides after treatment. Serum untargeted metabolomics intervention reduced ergothioneine, pentostatin, diadenosine tetraphosphate other metabolites modulated nicotinate nicotinamide metabolism, metabolic pathways, glyoxylate dicarboxylate pathways intestinal microorganisms. According to combined microbial metabolite analysis, influences modulating microbiota. summary, it was found treatment injury responses mice, disrupted host's flora, affected changes flora metabolites, thereby exacerbating development inflammation. Therefore, consumption nut can be associated with risk aggravating UC.

Ulcerative colitis (UC) is a chronic inflammatory disorder affecting the colon, with symptomatology influenced by factors including environmental, genomic, microbial, and immunological interactions. Gut microbiota dysbiosis, characterized bacterial population alterations, contributes to intestinal homeostasis disruption aberrant immune system activation, thereby exacerbating state. This study assesses therapeutic efficacy of intraperitoneal (IP) injected flavonoids (apigenin, luteolin, xanthohumol) in reduction parameters modulation gut murine model ulcerative colitis. Flavonoids interact modulating their composition serving as substrates for fermentation into other anti-inflammatory bioactive compounds. Our results demonstrate effectiveness luteolin xanthohumol treatment enhancing relative abundance microorganisms, attenuating pro-inflammatory species. Moreover, all three exhibit cytokine levels, strongly demonstrating utility alleviating associated physical UC symptoms. suggests that this molecule potential alternative or co-therapy conventional pharmacological interventions, potentially mitigating adverse effects. A limited impact on observed apigenin, attributed its solubility constraints via chosen administration route, resulting accumulation mesentery.

Inflammatory bowel disease (IBD) is a recurrent inflammatory condition affecting the gastrointestinal tract, and its clinical treatment remains suboptimal. Probiotics have shown effectiveness in alleviating dextran sulfate sodium salt (DSS)-induced colitis, exhibiting strain-specific anti-inflammatory properties. In this study, we compared therapeutic effects of five strains Bifidobacterium bifidum isolated from healthy adult feces on DSS-induced colitis mice. Additionally, investigated underlying mechanisms by examining gut microbiota composition microbial metabolome. Our findings highlighted superior efficacy B. M1-3 to other strains. It significantly improved symptoms, mitigated barrier disruption, reduced colonic inflammation DSS-treated Moreover, analysis revealed that increased abundance diversity microbiota. Specifically, it Muribaculaceae, Lactobacillus, Bacteroides, Enterorhabdus, while decreasing Escherichia-Shigella. Furthermore, our nontargeted metabolomics illustrated had regulatory effect various metabolic pathways, including tyrosine metabolism, lysine degradation, tryptophan metabolism. Importantly, confirmed efficiency was dependent These results are conducive development probiotic products for colitis.

Nevertheless, its confined predominantly human fecal sources. Following integration probiogenomics, significant advancements have been made safety assessment probiotics. However, adoption novel probiotic microorganisms has match requisite pace. Progress research concerning host-probiotic interactions by employing omics technologies, particularly animal models, notable. Nonetheless, comprehensive elucidation mechanisms action trial studies are lagging behind. Additionally, viability probiotics, spanning from production as foods transit colon, markedly improved through encapsulation techniques. opportunities exploration persist regarding alternative coating materials diverse methodologies.
