Abstract Advancing age is associated with chronic diseases which are the largest cause of death and disability in developed countries. With increasing life expectancy an ageing population, there a need to conduct trials extend healthy ageing, including targeting biological processes, prevent ageing-related diseases. The main objectives study as follows: (i) review outcome measures utilised focusing on pharmacological therapies, nutritional supplements medical devices; (ii) summarise views key stakeholders selection for trials. An analysis records from Clinicaltrials.gov database pertaining inception May 2022 was conducted. In addition, findings workshop attended by at annual UKSPINE conference were qualitatively analysed. Substantial heterogeneity found interventions evaluated outcomes included studies. Recruitment participants diverse backgrounds confounding effects multi-morbidity older adults identified challenges measuring participants. development core set can aid comparability across within different settings. provided important platform garner range perspectives this setting. Immune system aging is becoming a field of increasing public health interest because prolonged life expectancy, which not paralleled by an increase in expectancy. As age progresses, innate and adaptive immune systems undergo changes, are defined, respectively, as inflammaging senescence. A wealth available data demonstrates that these two conditions closely linked, leading to greater vulnerability elderly subjects viral, bacterial, opportunistic infections well lower post-vaccination protection. To face this novel scenario, in-depth assessment the players involved changing epidemiology demanded regarding individual concerted involvement cells mediators within endogenous exogenous factors co-morbidities. This review provides overall updated description changes affecting system, may be help understanding underlying mechanisms associated with main age-associated infectious diseases.

We analyze aging signatures of DNA methylation and longitudinal electronic medical records from the UK Biobank datasets observe that is driven by a large number independent infrequent transitions between metastable states in vast configuration space. The compound effect changes can be captured single stochastic variable, thermodynamic biological age (tBA), tracking entropy produced, hence information lost during aging. show tBA increases with age, causes linear irreversible drift physiological state variables, reduces resilience, drives exponential acceleration chronic disease incidence death risks. entropic character sets severe constraints on possibilities reversal. The most recognizable implications of tissue aging manifest themselves on the skin. Skin laxity, roughness, pigmentation disorders, age spots, wrinkles, telangiectasia or hair graying are symptoms physiological aging. Development senescent phenotype depends interaction between cells and remodeling skin’s extracellular matrix (ECM) that contains collagen elastic fiber. Aging changes occur due to combination both endogenous (gene mutation, cellular metabolism hormonal agents) exogenous factors (ultraviolet light, environmental pollutants, unsuitable diet). However, overproduction mitochondrial reactive oxygen species (ROS) is a key factor driving senescence. theories have disclosed range diverse molecular mechanisms associated with senescence body. Theories best supported by evidence include protein glycation, oxidative stress, telomere shortening, cell cycle arrest, limited number divisions. Accumulation ECM damage suggested be in skin Every indicates functional morphological change may used as biomarker Senescence-associated β -galactosidase (SA-β-gal), inhibitors (p16INK4a, p21CIP1, p27, p53), DNA segments chromatin alterations reinforcing (DNA-SCARS), senescence-associated heterochromatin foci (SAHF), shortening telomeres downregulation lamina B1 constitute just an example biomarkers known so far. also assessed non-invasively through measuring fluorescence advanced glycation end-products (AGEs). This review summarizes recent knowledge pathogenesis clinical conditions well

Abstract Possible sustainable resources of beneficial compounds for various applications are agricultural crop and food production residues (ACFPR), which supported by considerable efforts to characterize their compositions biological activities. This knowledge can be utilized the rational selection residue extracts components possible use in development value‐added products, such as anti‐ageing cosmetics. The appearance wrinkles, pigmentation, a reduction skin elasticity typical signs ageing that often alleviated natural product‐based preparations. Here, we silico approaches identify from with potential alleviate symptoms or reverse process. Target predictions combined extensive database literature searches were present ACR proteins linked ageing. binding affinity products selected using molecular docking respective intermolecular interaction analyses predicted provide an indicative measure compounds' activity. A number interact protein targets collagenase, elastase, hyaluronidase identified. In addition findings, cytotoxicity assays conducted rose hip seed against Hs294T (human metastatic melanoma, ATCC HTB‐140) MRC‐5 (normal fetal lung fibroblasts, CCL 171), demonstrating selective cytotoxicity. ELISA revealed induced significant increase SIRT1 levels (160% control) TGF‐β (80% control). These experimental results support modulating key involved ageing, reinforcing viability ingredients cosmetic formulations. An analysis relationships pathways organic substances sources affect offers developing formulations rejuvenation synergistic effects utilizing design innovative skincare laying framework more effective screening different applications.
