Reduced glutathione (GSH) is the most abundant non-protein endogenous thiol. It a ubiquitous molecule produced in organs, but its synthesis predominantly liver, tissue charge of storing and distributing it. GSH involved detoxification free radicals, peroxides xenobiotics (drugs, pollutants, carcinogens, etc.), protects biological membranes from lipid peroxidation, an important regulator cell homeostasis, since it participates signaling redox, regulation degradation proteins (S-glutathionylation), signal transduction, various apoptotic processes, gene expression, proliferation, DNA RNA synthesis, etc. transport vital step cellular homeostasis supported by liver through providing extrahepatic organs (such as kidney, lung, intestine, brain, among others) with said antioxidant. The wide range functions within which shows that glutathione’s role goes beyond being simple antioxidant agent; therefore, importance this tripeptide needs to be reassessed broader metabolic perspective.

Abstract Background: N-acetylcysteine (NAC) is a mucolytic agents with anti-inflammatory properties that has been suggested as an adjunctive therapy in patients COVID-19 pneumonia. Objectives: We conducted systematic review and meta-analysis to evaluate available evidence on the possible beneficial effects of NAC SARS-CoV-2 infection. Methods: In September 2022, we comprehensive search Pubmed/Medline Embase randomized controlled trials (RCTs) observational studies Study selection, data extraction risk bias assessment was performed by two independent authors. RCTs were analyzed separately. Results: included 3 5 non-randomized efficacy COVID-19, enrolling 315 20826 respectively. Regarding in-hospital mortality, summary effect all OR: 0.85 (95% CI: 0.43 1.67, I 2 =0%) for 1.02 0.47 2.23, =91%). Need ICU admission only reported 1 RCT (OR: 0.86, 95% CI:0.44-1.69, p=0.66), while need invasive ventilation (OR:0.91, CI:0.54 1.53, =0). Risk low RCTs, but certainty very outcomes due serious imprecision indirectness. Conclusion: The low, thus recommendations clinical practice cannot be yet made. For hard point estimates are close line no effect, have high degree heterogeneity some them suggesting favorable results receiving NAC. More research warranted insure both effective safe

Background Multisystem inflammatory syndrome in children (MIS-C) is a rare and severe post-COVID-19 complication with multiple phenotypes. Objectives The aim of this study to biomarkers (cytokines oxidative stress) critical MIS-C patients observe if there association between these mortality. Methods A single-center prospective enrolled (with positive molecular test), aged 1 month 18 years age. Data was collected from 20 pediatric intensive care unit (PICU)’s bed. Inflammatory stress markers) were performed on day 3 after hospitalization. Survival rate calculated, Kaplan-Meier curves plotted. Univariate multivariate Cox regression analyses conducted. ROC (Receiver Operating Characteristic) curve analysis performed. Results conclusions total 41 out 109 admitted at PICU suspected during the period included, which 33 (80.5%) male, 9 (22%) under one year old, 30 (73.2%) presented comorbidities. Among them, 16 (39%) did not survive. mean survival time shorter higher levels IL-17A (≥ 19.71 pg/mL) (115 vs 323 days, p = 0.004). Higher associated mortality both crude model (HR 1.03, CI95% 1.004-1.057, 0.022) adjusted 1.043, 1.013-1.075, 0.012). revealed cut-off value for 14.32 pg/ml. other immunological markers demonstrate an (p>0.05). Our findings suggest that high are greater risk death.

We focused on evaluating oxidative stress as a major mechanism of cell damage in patients with COVID-19 infection by simultaneously assessing standard biomarkers vivo—for the very first time this specific combination—alongside typical clinical inflammation. Standard were used to evaluate status and antioxidant activity blood plasma healthy controls. These included TBARSs (Thiobarbituric Acid-Reactive Substances), SOD (Super Oxide Dismutase), CAT (catalase), GRA (glutathione reductase) activities, AOC (antioxidant capacity). All inflammation data confirmed highly activated immune response tested patients: WBCs (white cells) increased nearly 100%, LYMs (lymphocytes) ~30%, CRP (C-reactive protein) rose over 2200%, ESR (erythrocyte sedimentation rate) ~320% compared established maximum control levels. The results that involved free-radical-mediated mechanism: TBARS levels almost 3-fold, decreased more than 4-fold, was 5-fold, 1.4 times, suppressed 2.5 times. associated activity. these changes contribute severity disease, complications, mortality patients.

Inflammasome activation is one of the first steps in initiating innate immune responses. In this work, we studied inflammasomes airways critically ill COVID-19 patients and effects N-acetylcysteine (NAC) on inflammasomes. Tracheal biopsies were obtained from without no respiratory disease (control, n = 32), SARS-CoV-2 B.1 variant (n 31), B.1.1.7 VOC alpha 20) patients. Gene expression protein measured by RT-qPCR immunohistochemistry. Macrophages bronchial epithelial cells stimulated with different S, E, M, N recombinant proteins presence or absence NAC. NLRP3 inflammasome complex was over-expressed activated associated systemic inflammation 28-day mortality. TLR2/MyD88 redox NOX4/Nrf2 ratio also variant. The combination S-E-M increased cytokine release macrophages through TLR2. NAC inhibited mosaic (S-E-M)-induced activation. summary, over-activated severe addition, can reduce induced vitro, which may be potential translational value

Proinflammatory bioactive lipid mediators and oxidative stress are increased in coronavirus disease 2019 (COVID-19). The randomized controlled single-blind trial COVID-Omega-F showed that intravenous omega-3 polyunsaturated fatty acids (n-3 PUFA) shifted the plasma signature of COVID-19 towards proresolving precursor levels decreased leukotoxin diols, associated with a beneficial immunodulatory response. present study aimed to determine effects n-3 PUFA on urinary oxylipidome COVID-19. From trial, 20 patients hospitalized for had available serial samples collected at baseline, after 24-48 h, completing 5 days treatment one daily infusion (2 mL/kg) either placebo (NaCl; n = 10) or emulsion containing 10 g per 100 mL (n 10). Urinary eicosanoids isoprostanes were analyzed by liquid chromatography coupled tandem mass spectrometry (LC-MS/MS). Erythrocytes obtained different time-points from used determination reactive oxygen species. Intravenous administration altered eicosanoid metabolites inflammation thrombosis, endothelial function mediator prostacyclin. Furthermore, non-enzymatic metabolism was skewed PUFA-derived potential anti-inflammatory pro-resolving effects. marker 15-F2t-isoprostane significantly lower receiving treatment, who also exhibited erythrocyte compared placebo-treated patients. These findings point additional through oxylipin profile
