Bone defects caused by trauma, tumor resection, and infections are significant clinical challenges. Excessive reactive oxygen species (ROS) usually accumulate in the defect area, which may impair function of cells involved bone formation, posing a serious challenge for repair. Due to potent ROS scavenging ability, as well potential anti-inflammatory immunomodulatory activities, antioxidants play an indispensable role maintenance protection health have gained increasing attention recent years. This narrative review aims give overview main research directions on application antioxidant compounds repair over past decade. In addition, positive effects various their biomaterial delivery systems summarized provide new insights exploring antioxidant-based strategies

Placentation is a key and tightly regulated process that ensures the normal development of placenta fetal growth. Preeclampsia (PE) hypertensive pregnancy-related disorder involving about 5–8% all pregnancies clinically characterized by de novo maternal hypertension proteinuria. In addition, PE are also increased oxidative stress inflammation. The NRF2/KEAP1 signaling pathway plays an important role in protecting cells against damage due to reactive oxygen species (ROS) levels. ROS activate NRF2, allowing its binding antioxidant response element (ARE) region present promoter several genes such as heme oxygenase, catalase, glutathione peroxidase superoxide dismutase neutralize ROS, damages. this review, we analyze current literature regarding preeclamptic pregnancies, discussing main cellular modulators pathway. Moreover, discuss natural synthetic compounds can regulate vivo vitro models.

Osteoporosis, a metabolic bone disease characterized by low mineral density and deterioration of microarchitecture, has led to high risk fatal osteoporotic fractures worldwide. Accumulating evidence revealed that sexual dimorphism is notable feature osteoporosis, with sex-specific differences in epidemiology pathogenesis. Specifically, females are more susceptible than males while prone disability or death from the disease. To date, sex chromosome abnormalities steroid hormones have been proven contribute greatly osteoporosis regulating functions cells. Understanding its related complications essential for improving treatment strategies tailored women men. This literature review focuses on mechanisms underlying mainly population aging patients, chronic glucocorticoid administration, diabetes. Moreover, we highlight implications developing therapeutics preventive screening approaches Additionally, challenges translating bench research bedside treatments future directions overcome these obstacles will be discussed.

Maejo 341 Sweet potato (MSP) is a new purple sweet variety cultivated in Northern Thailand, but its health benefits are unknown. This study aimed to investigate antioxidant, anti-inflammatory, and anti-osteoporotic activities, as well anthocyanin content. The peel flesh of MSP were extracted with ethanol water. Compared the extracts, extracts presented greater antioxidant capacity rich phenolics, flavonoids, anthocyanins, namely, cyanidin-3-O-glucoside, peonidin-3-O-glucoside, pelargonidin-3-O-glucoside, cyanidin, peonidin. suppressed lipopolysaccharide-induced inflammation by inhibiting secretion proinflammatory cytokines enzymes, including TNF-α, IL-1β, IL-6, COX-2, iNOS, reducing nitric oxide matrix metalloproteinase-9 secretion. inhibited RANKL-induced NF-κB MAPK pathways downregulated osteoclastogenic marker expression. Under LPS RANKL treatment, notably reduced reactive oxygen species production while increasing gene Furthermore, they increased osteoblast viability slightly raise alkaline phosphatase activity. These findings suggest that could be used functional food alleviate oxidative stress inflammation-related osteoporosis.

A metabolic bone disorder called osteoporosis is characterized by decreased mass and compromised microarchitecture. This condition can deteriorate bones raise the risk of fractures. The two main causes are an increase in osteoclast activity or quantity a decrease osteoblast viability. Numerous mechanisms, including estrogen shortage, aging, chemical agents, mechanical loads, have been linked to osteoporosis. Inflammation oxidative stress recently osteoporosis, according increasing number studies. primary medications used treat at moment bisphosphonates selective receptor modulators (SERMs). These work well for brought on aging deprivation, however, they do not target inflammation stress-induced In addition, these drugs some limitations that attributed various side effects overcome. Traditional Chinese medicine (TCM) has applied many years high safety profile. Therefore, this review, literature related botanical effect was searched for. Moreover, pharmacologically active ingredients herbs pathways were discussed may contribute discovery more safe effective treatment

Abstract Osteoporosis (OP) can result in slower bone regeneration than the normal condition due to abnormal oxidative stress and high levels of reactive oxygen species (ROS), a detrimental for formation, making OP‐related healing significant clinical challenge. As osteogenic differentiation ability marrow mesenchymal stem cells (BMSCs) is closely related regeneration; currently, this study assessed effects Picein on BMSCs vitro osteoporotic defect vivo. Cell viability was determined by CCK‐8 assay. The production malonaldehyde, superoxide dismutase activities, glutathione evaluated using commercially available kits, flow cytometry analysis adopted detect macrophage polarization. Osteogenic capacity alkaline phosphatase (ALP) activity, ALP staining, Alizarin red S staining. expression osteogenic‐related proteins (OPN, Runx‐2, OCN) genes (ALP, BMP‐4, COL‐1, Osterix) were Western blotting real‐time PCR (RT‐PCR). In addition, proliferation, migration ability, angiogenic human umbilical vein endothelial (HUVECs) EdU scratch test, transwell assay, tube formation respectively. Angiogenic‐related (VEGF, vWF, CD31) also RT‐PCR. Results showed that alleviated erastin‐induced stress, enhanced BMSCs, angiogenesis HUVECs, protects against ferroptosis through Nrf2/HO‐1/GPX4 axis. Moreover, regulate immune microenvironment promoting polarization M2 macrophages vitro. reduce inflammation promotes OP rat models Altogether, these results suggested promote alleviate via pathway, offering as novel antioxidant agent treating defect.

The prolonged use of exogenous glucocorticoids, such as dexamethasone (Dex), is the most prevalent secondary cause osteoporosis, known glucocorticoid-induced osteoporosis (GIO). current study examined preventative and synergistic effect aqueous chicory extract (ACE) ethanolic purslane (EPE) on GIO compared with Alendronate (ALN). phytochemical contents, elemental analysis, antioxidant scavenging activity, ACE EPE combination index were evaluated. Rats randomly divided into control, ACE, EPE, ACE/EPE MIX groups (100 mg/kg orally), Dex group (received 1.5 mg Dex/kg, Sc), four treated received MIX, ALN Dex. bone mineral density content, index, growth, turnover, oxidative stress measured. molecular analysis RANK/RANKL/OPG Nrf2/HO-1 pathways also causes by increasing stress, decreasing markers, reducing growth markers (OPG OCN), turnover resorption (NFATc1, RANKL, ACP, ALP, IL-6, TNF-α). In contrast, showed a prophylactic against Dex-induced modulating measured parameters histopathological architecture. conclusion, exerts powerful mode action different from ALN.
