Silent information regulator sirtuin 1 (SIRT1) is an NAD+-dependent deacetylase with potent anti-arterial aging activities. Its protective function in aging-related diseases has been extensively studied. In the microcirculation, SIRT1 plays a crucial role preventing microcirculatory endothelial senescence by suppressing inflammation and oxidative stress while promoting mitochondrial optimizing autophagy. It suppresses hypoxia-inducible factor-1α (HIF-1α)-mediated pathological angiogenesis healthy, physiological capillarization. As result, protects against microvascular dysfunction, such as diabetic microangiopathy, enhancing exercise-induced skeletal muscle capillarization energy metabolism. brain, upregulates tight junction proteins strengthens their interactions, thus maintaining integrity of blood-brain barrier. The present review summarizes recent findings on regulation SIRT1, underlying mechanisms, various approaches to modulate activity microcirculation. importance molecular target diseases, retinopathy stroke, underscored, along need for more clinical evidence support modulation

Aging is linked to a time-associated decline in both cellular function and repair capacity leading malfunction on an organismal level, increased frailty, higher incidence of diseases, death. As the population grows older, there need reveal mechanisms associated with aging that could spearhead treatments postpone onset age-associated decline, extend healthspan lifespan. One possibility targeting sirtuin SIRT1, founding member family, highly conserved family histone deacetylases have been metabolism, stress response, protein synthesis, genomic instability, neurodegeneration, DNA damage repair, inflammation. Importantly, sirtuins also implicated promote health lifespan extension, while their dysregulation has cancer, neurological processes, heart disorders. Resveratrol is a natural polyphenol (stilbenoid), which can be found in grape skin, red wine, blueberries, peanuts and others. The biological properties of resveratrol, particular antioxidant, anti-inflammatory, anticancer, estrogenic, vasorelaxant cardioprotective activity, are the main reason for its importance medicine pharmacy. Despite all advantages, however, there many problems related to this polyphenolic substance, such as low stability, water insolubility, poor bioavailability fast metabolism. For reason, scientists currently searching different approaches dealing with these problematic improving therapeutic usage resveratrol. This review summarizes mechanisms effects determined vitro vivo, limitations drug. article emphasizes new improvement resveratrol delivery, nanoencapsulation, formation nanocrystals, prodrugs structure analogues.

Resveratrol, a polyphenolic compound known for its diverse biological activities, has demonstrated multiple pharmacological effects, including anti-inflammatory, anti-aging, anti-diabetic, anti-cancer, and cardiovascular protective properties. Recent studies suggest that these effects are partly mediated through the regulation of macrophage polarization, wherein macrophages differentiate into pro-inflammatory M1 or anti-inflammatory M2 phenotypes. Our review highlights how resveratrol modulates polarization various signaling pathways to achieve therapeutic effects. For example, can activate senescence-associated secretory phenotype (SASP) pathway inhibit signal transducer activator transcription (STAT3) sphingosine-1-phosphate (S1P)-YAP axes, promoting suppressing thereby inhibiting tumor growth. Conversely, it promote suppress by NF-κB activating PI3K/Akt AMP-activated protein kinase (AMPK) pathways, thus alleviating inflammatory responses. Notably, effect on is concentration-dependent; moderate concentrations tend while higher may favor polarization. This concentration dependence offers new perspectives clinical treatment but also underscores necessity precise dosage control when using resveratrol. In summary, exhibits significant potential in regulating treating related diseases.

Age-related diseases have emerged as a global concern the population ages. Consequently, understanding underlying causes of aging and exploring potential anti-aging interventions is imperative. In females, ovaries serve principal organs responsible for ovulation production female hormones. The are related to infertility, menopause, associated menopausal syndromes, with menopause representing culmination ovarian aging. Current evidence indicates that may contribute dysfunction across multiple organ systems, including, but not limited to, cognitive impairment, osteoporosis, cardiovascular disease. Nevertheless, due widespread distribution sex hormone receptors throughout body, affects only these specific also influences broader spectrum age-related in women. Despite this, impact on overall has been largely neglected. This review provides thorough summary diseases, encompassing nervous, circulatory, locomotor, urinary, digestive, respiratory, endocrine systems. Additionally, we outlined prospective therapeutic approaches addressing both aim mitigating their impacts preserving women's fertility, physical health, psychological well-being.
