Comorbid chronic neuropathic pain and anxiety is a common disease that represents major clinical challenge. The underlying mechanisms of are not entirely understood, which limits the exploration effective treatment methods. Glutamatergic neurons in ventrolateral periaqueductal gray (vlPAG) have been implicated regulating pain, but potential roles vlPAG pain-induced investigated. Herein, whole-cell recording immunofluorescence showed excitability CamkIIα (vlPAG CamkIIα+ neurons) was decreased mice with spared nerve injury (SNI), while electroacupuncture (EA) activated these neurons. We also chemogenetic inhibition resulted allodynia anxiety-like behaviors naive mice. Furthermore, activation reduced SNI, EA had similar effect alleviating symptoms. Nevertheless, combined failed to further relieve behaviors. Artificial abolished analgesic anxiolytic effects EA. Overall, our study reveals novel mechanism shows may comorbid by activating

Background and objective Whilst acupuncture is widely used for treating psychosomatic diseases, there little high-quality evidence supporting its application in comorbid perimenopausal depression (PMD) insomnia (PMI) which are common complaints during climacteric. This feasibility, patient-assessor-blinded, randomized, sham-controlled clinical trial addresses this gap by investigating the efficacy safety of on depressed mood poor sleep women with PMD PMI. Methods Seventy eligible participants were randomly assigned to either real-acupuncture (RA) or sham-acupuncture (SA) groups. Either RA SA treatment delivered 17 sessions over 8 weeks. The primary outcomes changes 17-items Hamilton Depression Rating Scale (HAM-D ) Pittsburgh Sleep Quality Index (PSQI) scores, from baseline 16-week follow-up. Secondary outcome measures involved anxiety symptoms, quality life, participants' experience satisfaction treatment. Blood samples taken measure reproductive hormone levels. Intention-To-Treat Per-Protocol analyses conducted linear mixed-effects models. James' Bang's blinding indices assess adequacy blinding. Results Sixty-five completed all sessions, 54 41 eight- follow-ups, respectively. At post-treatment 8-week follow-up, group showed a significantly greater reduction PSQI scores than did; although HAM-D was significant, change not statistically different that SA. There no significant mean differences between follow-up group. between-group serum All treatments tolerable serious adverse events reported, successful. Conclusion Acupuncture safe can contribute clinically relevant improvements PMI, satisfactory short-and medium-term effects. Whether anti-depressive benefit specific non-specific remains be determined. No found any longer-term compared sham at 16 Further research required elucidate mechanisms underlying short medium term effects acupuncture.

Acupuncture, used as an adjunctive therapy anti-Parkinson medications, has shown positive effects PD. However, high-quality clinical evidence to support the effectiveness acupuncture for PD poor is lacking. Objective To assess safety efficacy real (RA) vs sham (SA) who have quality. Design, Setting, Participants This single-center randomized trial was performed at The First Affiliated Hospital Guangzhou University Chinese Medicine China from February 18, 2022, 2023. Male Sprague-Dawley (SD) rats were randomly divided into control group (CON), chronic unpredictable mild (CUMS) group, CUMS + electroacupuncture (EA), fluoxetine (FLX) (n = 10/group). Abstract Study Objectives Chronic insomnia disorder (CID) is a prevalent sleep that frequently cooccurs with anxiety. The association between and anxiety has been established; however, the neurobiological basis of this relationship remains unclear. This study aimed to investigate neural markers CID patients without determine whether specific connectivity mediates Methods included 180 participants, comprising (CID-A), (CID-NA), good controls. All participants completed self-reported measures quality severity underwent functional magnetic resonance imaging. Brain integration was measured using density (FCD) resting-state (rsFC). Correlation mediation analyses were used examine relationships among brain connectivity, quality, severity. Results CID-NA CID-A groups showed decreased local FCD in medial prefrontal cortex (mPFC) disrupted rsFC precuneus other regions. Only group exhibited altered long-range anterior default mode network (DMN, e.g. mPFC) posterior DMN (e.g. precuneus). Mediation analysis revealed dysconnectivity underlying poor symptoms. Conclusions identified shared distinct circuit disruptions groups, deficits as potential mechanism through which augments These findings may facilitate development personalized therapies for associated problems.

Depression is commonly associated with gastrointestinal (GI) disorders, such as constipation, which can potentially intensify depressive symptoms. The interplay between these conditions believed to be facilitated by the gut-brain axis, suggests a complex bidirectional interaction. Current treatments, antidepressants and prokinetics, are often side effects high recurrence rates, highlighting need for effective treatments targeting both depression constipation. This study was designed assess therapeutic efficacy of electroacupuncture (EA) in conjunction Tongbian decoction (TB) management while also investigating underlying mechanisms through lens axis. Chronic unpredictable mild stress (CUMS) employed induce comorbidity model constipation mice, followed administration EA, EA + TB, fluoxetine (FLX). findings demonstrated that antidepressant combination were more pronounced than those alone.
