As the circadian clock regulates fundamental biological processes, disrupted clocks are often observed in patients and diseased tissues. Determining time of patient or tissue focus is essential medicine research. Here we present tauFisher, a computational pipeline that accurately predicts from single transcriptomic sample by finding correlations between rhythmic genes within sample. We demonstrate tauFisher's performance adding timestamps to both bulk single-cell samples collected multiple types experimental settings. Application tauFisher at cell-type level RNAseq dataset mouse dermal skin implies greater phase heterogeneity may explain dampened rhythm collective core gene expression immune cells compared fibroblasts. Circadian rhythms are intrinsic 24-hour oscillations found in nearly all life forms. They orchestrate key physiological and behavioral processes, allowing anticipation response to daily environmental changes. These manifest across entire organisms, various organs, through intricate molecular feedback loops that govern cellular oscillations. Recent studies describe circadian regulation of pathogens, including parasites, bacteria, viruses, fungi, some which have their own while others influenced by the rhythmic environment hosts. Pathogens target specific tissues organs within host optimize replication. Diverse compositions interplay among cell types create unique microenvironments different tissues, distinctive biology. Hence, residing pathogens exposed cyclic conditions, can profoundly impact host–pathogen interactions. This review explores influence mammalian tissue-specific interactions on dynamics pathogen–host relationships. Overall, this demonstrates between body’s internal timekeeping system its susceptibility has implications for future infectious disease research treatment.

Abstract First described in the early 20th century, diurnal oscillations stem cell proliferation exist multiple internal epithelia, including gastrointestinal tract, and epidermis. In mouse epidermis, 3- to 4-fold more cells are S-phase during night than day. More recent work showed that an intact circadian clock intrinsic keratinocytes is required for these epidermal proliferation. Cutaneous pigmentation was recently shown to be an event regulated by clock proteins. Cryptochrome (CRY) is a key protein composing the feedback loop of circadian clock, however, function CRY in melanocytes remains unclear. Here, we found that KL001, synthetic small molecule modulator CRY1, inhibited melanin synthesis, as well reduced melanocyte dendrite elongation and melanosome transport. In addition, dominant role CRY1 KL001-induced anti-melanogenesis revealed interfering RNA transfection. Cellular tyrosinase activity expression level melanogenic proteins, including tyrosinase, TRP-1, TRP-2, transport proteins like Rab27a, Cdc42 Myosin Va induced α-MSH were remarkably reversed after KL001 treatment. Mechanistically, activation melanogenesis through CREB-dependent downregulation MITF CREB phosphorylation mediated classical cAMP/PKA pathway. other activator, KL044 also suppressed cAMP/PKA/CREB pathway melanogenesis.
