Abstract: Wound healing is a sophisticated and orderly process of cellular interactions in which the body restores tissue architecture functionality following injury. Healing chronic diabetic wounds difficult due to impaired blood circulation, reduced immune response, disrupted repair mechanisms, are often associated with diabetes. Stem cell-derived extracellular vesicles (SC-EVs) hold regenerative potential, encapsulating diverse cargo proteins, RNAs, cytokines, presenting safe, bioactivity, less ethical issues than other treatments. SC-EVs orchestrate multiple processes by modulating communication, increasing angiogenesis, promoting recruitment differentiation progenitor cells, thereby potentiating reparative milieu for wound healing. Therefore, this review investigated effects mechanisms EVs from various stem cells healing, as well their limitations challenges. Continued exploration has potential revolutionize care. Abstract Stem cells (SCs) have been used therapeutically for decades, yet their applications are limited by factors such as the risk of immune rejection and potential tumorigenicity. Extracellular vesicles (EVs), a key paracrine component stem cell potency, overcome drawbacks cell-free therapeutic agent play an important role in treating various diseases. However, EVs derived from two-dimensional (2D) planar culture SCs low yield face challenges large-scale production, which hinders clinical translation EVs. Three-dimensional (3D) culture, given its ability to more realistically simulate vivo environment, can not only expand large quantities, but also improve activity EVs, changing content improving effects. In this review, we briefly describe advantages EV-related applications, provide overview 3D finally focus on specific future perspectives different SCs. Graphical

Abstract Background The use of stem cell-derived exosomes (Exos) as therapeutic vehicles is receiving increasing attention. Exosome administration has several advantages over cell transplantation, thus making promising candidates for large-scale clinical implementation and commercialization. However, exosome extraction purification efficiencies are relatively low, heterogeneity high due to differences in culture conditions viability. Therefore, this study, we investigated a priming procedure enhance the production effects from human umbilical cord mesenchymal cells (hucMSCs). After preconditioning hucMSCs with agonists/inhibitors that target Wnt/β-catenin pathway, assessed both efficacy optimized context diabetic wound healing, hoping provide safer, more stable effective option application. Results Wnt signalling pathway agonist CHIR99021 increased by 1.5-fold without causing obvious changes characteristics or size particles. Further studies showed promoted facilitating exocytosis. Extracellular vesicles (EVs) derived from human organoids are phospholipid bilayer-bound nanoparticles that carry therapeutic cargo. However, the low yield of EVs remains a critical bottleneck for clinical translation. Vertical-Wheel bioreactors (VWBRs), with unique design features, facilitate scalable production secreted by blood vessel (BVOs) under controlled shear stress, using aggregate- and microcarrier-based culture systems. Human induced pluripotent stem cell-derived BVOs cultured as aggregates or on Synthemax II microcarriers within VWBRs (40 80 rpm) were compared to static controls. The characterized metabolite profiling, flow cytometry, gene expression EV biogenesis markers. nanoparticle tracking analysis, electron microscopy, Western blotting. Lipidomics provided insights into lipid composition, while functional assays assessed impact in D-galactose-induced senescence model. VWBR cultures showed more aerobic metabolism higher genes control. different conditions comparable size, but yields significantly microcarrier dynamic than aggregates. Lipidomic profiling revealed minimal variation (< 0.36%) total content; however, distinct differences identified chain lengths saturation levels, affecting key pathways such sphingolipid neurotrophin signaling. BVO demonstrated abilities reducing oxidative stress increasing cell proliferation vitro. differentiated (in particular 40 produce 2-3 fold (per mL) bio manufactured have exosomal characteristics cargo, showing properties vitro assays.
