Abstract Curcumin, a natural bioactive polyphenol with diverse molecular targets, is well known for its anti‐oxidation and anti‐inflammatory potential. However, curcumin exhibits low solubility (<1 µg mL −1 ), poor tissue‐targeting ability, rapid oxidative degradation, resulting in bioavailability stability inflammatory therapy. Here, poly(diselenide‐oxalate‐curcumin) nanoparticle (SeOC‐NP) dual‐reactive oxygen species (ROS) sensitive chemical moieties (diselenide peroxalate ester bonds) fabricated by one‐step synthetic strategy. The results confirmed that dual‐ROS endowed SeOC‐NP the ability of targeted delivery significantly suppress degradation high‐efficiency In detail, amount SeOC about 4‐fold lower than free an microenvironment. As result, enhanced antioxidant activity efficacy vitro analysis scavenging intracellular ROS suppressing secretion nitric oxide pro‐inflammatory cytokines. mouse colitis models, orally administered can remarkably alleviate symptoms IBD maintain homeostasis gut microbiota. Abstract Background Herbal nanoparticles are made from natural herbs/medicinal plants, their extracts, or a combination with other nanoparticle carriers. Compared to traditional herbs, herbal lead improved bioavailability, enhanced stability, and reduced toxicity. Previous research indicates that medicine nanomaterials rapidly advancing making significant progress; however, bibliometric analysis knowledge mapping for currently lacking. We performed by retrieving publications related the Web of Science Core Collection (WoSCC) database spanning 2004 2023. Data processing was using R package Bibliometrix, VOSviewers, CiteSpace. Results In total, 1876 articles were identified, originating various countries, China being primary contributing country. The number in this field increases annually. Beijing University Chinese Medicine, Shanghai Traditional Saveetha India prominent institutions domain. Abstract Treatment for inflammatory bowel disease (IBD) is challenging since current anti-inflammatory and immunosuppressive therapies do not address the underlying causes of illness, which include increased levels reactive oxygen species (ROS) dysbiosis gut commensal microbiota. Additionally, these treatments often have systemic off-target effects adverse side effects. In this study, we developed a prebiotic yeast β-glucan nanocomplex coated with bio-adhesive polydopamine (YBNs@PDA) to effectively prolong their retention time in gastrointestinal (GI) tract. The oral administration YBNs@PDA restored epithelium barriers, reduced ROS levels, minimized drug exposure while improved therapeutic efficacy an acute colitis mouse model. Furthermore, 16S ribosomal RNA genes sequencing demonstrated higher richness diversity microflora composition following treatments. particular, markedly augmented abundance Lachnospiraceae NK4A136 Bifidobacterium , both are probiotics crucial roles relieving via retaining homeostasis. Cumulatively, results demonstrate that potential as novel drug-free, ROS-scavenging microbiota regulation nanoplatform treatment GI disorders.

Carrier-free nanodrugs with extraordinary active pharmaceutical ingredient (API) loading (even 100%), avoidable carrier-induced toxicity, and simple synthetic procedures are considered as one of the most promising candidates for disease theranostics. Substantial studies commercial success "carrier-free" nanocrystals have demonstrated their strong clinical potential. However, practical translations remain challenging impeded by unpredictable assembly processes, insufficient delivery efficiency, an unclear in vivo fate. In this Perspective, we systematically outline contemporary emerging carrier-free based on diverse APIs, well highlight opportunities challenges translation. Looking ahead, further improvements design preparation, drug delivery, efficacy, safety nanomedicines essential to facilitate translation from bench bedside.
