Abstract Insufficient activation of the stimulator interferon genes (STING) signaling pathway and profoundly immunosuppressive microenvironment largely limits effect cancer immunotherapy. Herein, tumor (TME)‐responsive nanoparticles (PMM NPs) are exploited that simultaneously harness STING Toll‐like receptor 4 (TLR4) to augment via TLR4‐mediated nuclear factor‐kappa B stimulation, leading increased secretion type I interferons (i.e., 4.0‐fold enhancement IFN‐β) pro‐inflammatory cytokines promote a specific T cell immune response. Moreover, PMM NPs relieve immunosuppression TME by decreasing percentage regulatory cells, polarizing M2 macrophages M1 type, thus creating an immune‐supportive unleash cascade adaptive Combined with anti‐PD‐1 antibody, synergistic efficacy is achieved in both inflamed colorectal noninflamed metastatic breast models. rechallenging tumor‐free animals homotypic cells induced complete rejection, indicating generation systemic antitumor memory. These TME‐responsive may open new avenue achieve spatiotemporal orchestration activation, providing promising clinical candidate for next‐generation

Abstract Metals are an emerging topic in cancer immunotherapy that have shown great potential modulating immunity cycle and promoting antitumor by activating the intrinsic immunostimulatory mechanisms which been identified recent years. The main challenge of metal‐assisted lies fact free metals as ion forms easily cleared during circulation, even cause systemic metal toxicity due to off‐target effects. With rapid development nanomedicine, metal‐based smart nanosystems (MSNs) with unique controllable structure become one most promising delivery carriers solve issue, owing their various endogenous/external stimuli‐responsiveness release ions for metalloimmunotherapy. In this review, state‐of‐the‐art research progress metal‐related is comprehensively summarized. First, mainstream MSNs‐assisted will be delineated. immunological effects certain categorization MSNs different characters compositions then provided, followed representative exemplar applications treatment, synergistic combination immunotherapy. Finally, we conclude review a summary remaining challenges associated provide authors' perspective on further advances.

Abstract The introduction of glucose oxidase, exhibiting characteristics consumption and H 2 O production, represents an emerging antineoplastic therapeutic approach that disrupts nutrient supply promotes efficient generation reactive oxygen species (ROS). However, the instability natural enzymes their low efficacy significantly impede broader application. In this context, 2D Ca Mn 8 16 nanosheets (CMO NSs) designed engineered to serve as a high‐performance nanozyme, enhancing enzyodynamic effect for ferroptosis–apoptosis synergistic tumor therapy, are presented. addition mimicking activities glutathione peroxidase, catalase, CMO NSs exhibit oxidase‐mimicking activities. This feature contributes antitumor performance through cascade catalytic reactions, involving disruption supply, self‐supply , subsequent ROS generation. exogenous 2+ released from NSs, along with endogenous enrichment induced by peroxidase‐ collectively mediate overload, leading apoptosis. Importantly, ferroptosis process is triggered synchronously output consumption. application ultrasound stimulation further enhances efficiency treatment. work underscores crucial role in therapy against tumors.

Abstract With the economy's globalization and population's aging, cancer has become leading cause of death in most countries. While imposing a considerable burden on society, high morbidity mortality rates have continuously prompted researchers to develop new oncology treatment options. Anti-tumor regimens evolved from early single surgical combined (or not) chemoradiotherapy then current stage tumor immunotherapy. Tumor immunotherapy undoubtedly pulled some patients back death.
