Abstract The recent upswing in the integration of spatial multi-omics for conducting multidimensional information measurements is opening a new chapter biological research. Mapping landscape various biomolecules including metabolites, proteins, nucleic acids, etc., and even deciphering their functional interactions pathways believed to provide more holistic nuanced exploration molecular intricacies within living systems. Abstract The analysis of low molecular weight (LMW) compounds is great interest to detect small pharmaceutical drugs rapidly and sensitively, or trace understand metabolic pathways. Matrix‐assisted laser desorption/ionization mass spectrometry (MALDI MS) plays a central role in the high (bio)molecules. However, its application for LMW restricted by spectral interferences m/z region, which are produced conventional organic matrices. Several strategies regarding sample preparation have been investigated overcome this problem. A different rationale centred on developing new matrices not only meet fundamental requirements good absorption ionization efficiency, but also vacuum stable “MALDI silent”, i. e., do give matrix‐related signals area. Matrix-assisted laser desorption and ionization (MALDI) is a widely used soft-ionization technique of modern mass spectrometry (MS). MALDI enables the analysis nearly all chemical compounds—including polar apolar (phospho)lipids—with minimum extent fragmentation. has some particular advantages (such as possibility to acquire spatially-resolved spectra) competitive with simultaneously developed ESI (electrospray ionization) MS. Although there are still methodological aspects that need be elucidated in more detail, it obvious careful selection an appropriate matrix plays most important role (lipid) analysis. Thyroid nodules (TNs) have emerged as the most prevalent endocrine disorder in China. Fine-needle aspiration (FNA) remains standard diagnostic method for assessing TN malignancy, although a majority of FNA results indicate benign conditions. Balancing accuracy while mitigating overdiagnosis patients with poses significant clinical challenge. Precise, noninvasive, and high-throughput screening methods high-risk diagnosis are highly desired but remain less explored. Developing such approaches can improve noninvasive like ultrasound imaging reduce nodule caused by invasive procedures. Herein, we investigate application gold-doped zirconium-based metal-organic framework (ZrMOF/Au) nanostructures metabolic profiling thyroid diseases. This approach enables efficient extraction urine metabolite fingerprints high throughput, low background noise, reproducibility. Utilizing partial least-squares discriminant analysis four machine learning models, including neural network (NN), random forest (RF), logistic regression (LR), support vector (SVM), achieved an enhanced (98.6%) discriminating cancer (TC) from low-risk TNs using panel. Through differences, potential pathway changes between malignancy identified. work explores rapid disease ZrMOF/Au-assisted LDI-MS platform, providing malignant tumors. Integrating this technologies enhance reliability tumor screening, helping to prevent unnecessary procedures reducing risk overtreatment nodules.
