The application of mechanical force to induce the formation and cleavage covalent bonds is a rapidly developing field within organic chemistry which has particular value in reducing or eliminating solvent usage, enhancing reaction rates also enabling preparation products are otherwise inaccessible under solution-phase conditions. Mechanochemistry found recent attention materials API formulation during rearrangement non-covalent interactions give rise functional products. However, this been known nucleic acids science almost since its inception late nineteenth century when Miescher exploited grinding facilitate disaggregation DNA from tightly bound proteins through selective denaturation latter. Despite wide ball milling amino acid chemistry, there have limited reports mechanochemical transformations involving nucleoside nucleotide substrates on preparative scales. A survey these reactions provided, majority used mixer mill display an universal requirement for liquid be present vessel. charged substrates, particular, provides considerable benefits both terms efficiency (reducing total processing times weeks hours) by minimising exposure aqueous conditions, access previously elusive materials. In absence large quantities heating, side-reactions can reduced eliminated. central contribution mechanochemistry (and specifically, milling) isolation biologically active derived nuclei will outlined. Abstract Mechanochemical activations in Rh III ‐ and Au I ‐catalyzed C−H alkynylations lead selectively to C 2 3 ‐alkynylated indoles. The processes show excellent functional group tolerance, do not require additional heating proceed under solventless conditions. Compared solvent‐based standard protocols, the reaction times are shorter catalyst quantities lower resulting high product yields ambient atmosphere mixer mills.
