Akt acts as a central protein influencing multiple pathologies in neurodegenerative diseases including AD and PD, using activators is promising management strategy. The current study characterized the effects of an Akt-activating peptide (Glu-Pro-Glu-Val-Leu-Pro, EPEVLR) obtained from walnut degradation on D-gal-induced memory impairment mice. EPEVLR was by hydrolysis proteins, identification sequences, screening for molecular docking sequentially. MWM test mice indicated that oral administration (80, 200 400 mg per kg day) significantly (p < 0.05) reversed impairment. WB tests mouse hippocampus confirmed could activate promoting its phosphorylation. In addition, further characterization (including TEM, ELISA, immunohistochemistry) related to phosphorylation showed lower Aβ p-tau levels, well more autophagosomes than those model group. Moreover, treatment increased Lactobacillus abundance reduced Helicobacter gut microbiome caused up-regulation SCFAs down-regulation LPS serum metabolites. Therefore, ingestion cognitive symptoms, possibly activation regulation intestinal flora pathway. Consumption EPEVLR-containing diet beneficial treating dysfunction.

Abstract The purpose of this article is to investigate the effects walnut ( Juglans regia L.) kernel pellicle on composition and properties enzymatic hydrolysis products meal using peptidomics bioinformatics. In study, a total 3423 peptide sequences were identified in peeled protein hydrolysates (PWPH) unpeeled (UWPH). Due presence pellicle, enzyme cleavage sites alkaline proteases precursor proteins altered, resulting differences number length peptides obtained. Principal component analysis indicates significant between PWPH UWPH. Combined with bioinformatics analysis, it was shown that peeling improved release peptides, formed more bioactive reduced allergenicity, water solubility. Seven acetylcholinesterase (AChE) inhibitory activity identified, Val‐Gly‐Ala‐Pro‐Phe‐Asp‐Gly‐Ala (VGAPFDGA) has strongest an IC 50 0.38 ± 0.01 mg/mL. These results confirmed could greatly change hydrolysates, seven novel reported showed AChE activity.

Exosome-like nanovesicles (ELNs) derived from plants are nanoscale vesicles isolated edible plant sources. Lycium ruthenicum Murray (LRM) has garnered growing attention for its dietary value and therapeutic benefits. In this study, a PEG6000-based method was developed to isolate LRM-ELNs. Response surface methodology (RSM) used optimize the extraction conditions obtain optimal efficiency. When PEG6000 concentration at 11.93%, relative centrifugal force 9720 g, incubation time 21.12 h, maximum LRM-ELN yield 4.24 g/kg. This optimization process yielded LRM-ELNs with particle size of 114.1 nm charge −6.36 mV. Additionally, mitigated Aβ-induced apoptosis in HT22 cells by enhancing mitochondrial membrane potential (MMP), lowering Bax/Bcl-2 ratio, reducing Cleaved Caspase-3 expression. Furthermore, alleviated oxidative stress promoting nuclear translocation Nrf2 upregulating expression HO-1 NQO1. These findings indicate that exert protective effects against damage may be considered as supplement Alzheimer’s disease prevention.
