Abstract The analysis of extracellular vesicles (EV) in blood samples is under intense investigation and holds the potential to deliver clinically meaningful biomarkers for health disease. Technical variation must be minimized confidently assess EV‐associated biomarkers, but impact pre‐analytics on EV characteristics remains minimally explored. We present results from first large‐scale Blood Benchmarking (EVBB) study which we systematically compared 11 collection tubes (BCT; six preservation five non‐preservation) three processing intervals (BPI; 1, 8 72 h) defined performance metrics ( n = 9). EVBB identifies a significant multiple BCT BPI diverse set reflecting sample quality, ex‐vivo generation blood‐cell derived EV, recovery molecular signatures. assist informed selection optimal analysis. proposed serve as framework guide future research further support methodological standardization studies.
