In this study, we aimed to evaluate the anti-inflammatory and anti-apoptotic effects of melatonin (MLT) on neuropathic pain (NP)-induced anxiety depression in a rat model. Adult male rats were separated into four groups, i.e., Sham-VEH: healthy animals received vehicle, Sham-MLT (10 mg/kg), chronic constrictive injury (CCI)-VEH: nerve ligation CCI-MLT. Next, used behavioral tests severity, anxiety, depression. Finally, sacrificed for molecular histopathological studies. Behavioral showed that NP could induce depressive- anxiety-like behaviors. activated NF-κB/NLRP3 inflammasome pathways by upregulating NF-κB, NLRP3, ASC, active Caspase-1, also enhancing concentrations cytokines (IL-1β IL-18) prefrontal cortex (PFC) hippocampus (HC). upregulated Bax, downregulated Bcl2, increased cell apoptosis HC PFC. The treated with MLT eliminated NP, as reduced improved anxiety- depressive-like behaviors, ameliorated pathways, modulated levels promote survival from modulating Bax Bcl2. Therefore, it might be inferred its properties mediate beneficial NP-induced affective disorders.
