Antimicrobial resistance is a critical global challenge in the 21st century, validating Sir Alexander Fleming's warning about misuse of antibiotics leading to resistant microbes. With dwindling arsenal effective antibiotics, it imperative concentrate on alternative antimicrobial strategies. Previous studies have not comprehensively discussed advantages and limitations various strategies, including bacteriophage therapy, probiotics, immunotherapies, photodynamic essential oils, nanoparticles peptides (AMPs) within single review. This review addresses that gap by providing an overview these non-antibiotic highlighting their pros cons, with particular emphasis (AMPs). We explore mechanism action AMPs against bacteria, viruses, fungi parasites. While hold significant promise, application mainstream drug development hindered challenges such as low bioavailability potential toxicity. However, advancements peptide engineering chemical modifications offer solutions enhance clinical utility. Additionally, this presents updates strategies aimed at improving cost, stability selective toxicity through peptidomimetics. These molecules demonstrated activity broad range pathogens, making them valuable candidates for integration into surface coatings prevent device-associated infections. Furthermore, we discuss approaches attaching functionalising surfaces. Finally, recommend comprehensive vivo evaluate efficacy mimetics, investigate synergistic combinations other assess medical devices.

In addition to the 20 canonical amino acids encoded in genetic code, there are two non-canonical ones: selenocysteine and pyrrolysine. The discovery of pyrrolysine synthetases (PylRSs) was a key event field code expansion research. importance this is mainly due fact that translation systems involving PylRS, tRNA (tRNAPyl) orthogonal endogenous organisms do not use acid protein synthesis. addition, belonging different groups also mutually orthogonal. This orthogonality based on structural features PylRS tRNAPyl, which include identical elements, such as condensed core, certain base pairs motifs tRNAPyl. suggests targeted changes these molecules enable their specificity for codon. Such modifications were successfully used obtain aaRS/tRNA allow incorporation unnatural into peptides. review presents results recent studies related correlation between structure activity tRNAPyl extend code.

Due to the extensive use of antibiotics, many highly resistant bacteria and extensively have been produced. In recent years, increase drug-resistant resulting proliferation increased incidence hospital-acquired infections caused great harm human health. Antimicrobial peptides (AMPs) are considered be an innovative antibiotic belong latest advances in this field. We designed a polypeptide verified its low minimum inhibitory concentration broad-spectrum activity against Gram-positive bacteria, Gram-negative fungi microbiology pharmacology. Several experiments confirmed that screened antimicrobial significant antidrug resistance also show therapeutic properties treatment systemic bacterial infections. addition, through our experimental research, it was proved antibacterial hydrogel composed poly(vinyl alcohol), sodium alginate, had excellent showed good wound healing ability.

The ongoing battle against viral pandemics continues, with the possibility of future outbreaks. search for effective antiviral compounds that can combat a diverse range viruses continues to be focal point research. This study investigated efficacy two natural antimicrobial peptides (AMPs) (lactoferricin and LL-37), synthetic AMPs (melimine Mel4), nine AMP mimics (758, 1091, 1096, 1083, 610, NAPL, 3-BIPL, 4-BIPL, Sau-22) influenza A virus strains H1N1 H3N2, human adenovirus 5 (HAdV-5), murine norovirus 1 (MNV-1). These were tested using pre-treatment, cell or post-cell entry treatment assays, electron microscopy, circular dichroism (CD), alongside evaluations cytotoxicity host cells. After 610 Sau-22 had relatively low IC50 values (2.35 6.93 µM, respectively) H3N2 (3.7 5.34 respectively). Conversely, not active these strains. For non-enveloped viruses, Mel4 mimic 1083 moderate activity HAdV-5 (Mel4 = 47.4 µM; 47.2 µM), whereas all AMPs, but none mimics, (LL-37 4.2 lactoferricin 23.18 melimine 4.8 8.6 µM). Transmission microscopy demonstrated targeted outer envelope while capsid viruses. CD showed adopted an α-helical structure in membrane mimetic environment, 758 remained unstructured. different groups is probably influenced by charge, hydrophobicity, size, and, case their secondary structure. findings underscore potential as promising candidates therapeutics both enveloped
