Diet is crucial for the prevention of colorectal cancer. Whole grains are source beneficial compounds this, such as fiber. The enrichment wholemeal rye bread with plant sterols (PSs) could increase its effects. This study aimed to assess potential antiproliferative effect this enriched food on colon adenocarcinoma cells (Caco-2) compared a non-enriched one. After human oral chewing, simulated semi-dynamic gastrointestinal digestion and colonic fermentation in simgi® system, liquids (FLs) obtained were used treatment cells. The carnivorous pitcher plants of the genus Nepenthes exhibit many ethnobotanical uses, including treatments stomachache and fever. Cell viability was through MTT analysis, whereas levels sphingosine-1-phosphate (S1P), sphingomyelins (SMs), ceramides (CERs) measured using LC-MS/MS. Phosphorylated 44/42 ERK1/2 NF-kB p65 (Ser536) protein by western blot analysis immunofluorescence staining. Apoptosis evaluated TUNEL staining flow cytometric assessment annexin-V propidium iodide (PI) labeling. Results: Treatment with significantly decreased cell survival, S1P, p-44/42 ERK1/2, p-NF-κBp65 cells compared controls. A substantial rise detected intracellular amounts C16-C24 CERs incubated 7-KSS.
