Bacterial keratitis is a serious ocular disease that affects millions of people worldwide each year, among which ≈25% are caused by Staphylococcus aureus. With the spread bacterial resistance, refractory methicillin-resistant S. aureus (MRSA) ≈120 000-190 000 annually and significant cause infectious blindness. Atomically precise gold nanoclusters (GNCs) recently emerged as promising antibacterial agents; although how GNC structure capping ligands control properties remains largely unexplored. In this study, adjusting ratio "bulky" thiol fragrance to linear zwitterionic ligand, conformation transformed from Au25 (SR)18 Au23 (SR)16 species, simultaneously converting both inactive into potent nanomaterials. Surprisingly, mixed-ligand capped GNCs exhibit superior potency compared their monoligand counterparts. The optimal highly against MRSA, showing >1024-fold lower minimum inhibitory concentration than corresponding free ligands. Moreover, it displays excellent in treating MRSA-induced mice with greatly accelerated corneal recovery (by approximately ninefold). Thus, study establishes feasible method synthesize ligand active non-antibacterial ligands, thereby increasing repertoires for combating multidrug-resistant infections.

Abstract Hyperglycemia exacerbates bacterial infections, disrupts angiogenesis, and perpetuates chronic inflammation in the wound. Therefore, effective management of diabetic wounds necessitates a synergistic approach that integrates local wound microenvironment remodeling with systemic hyperglycemia regulation. Herein, an unprecedented hierarchical cobalt pyrophosphate/DNA (Co 2 PPi/DNA) nanocomposite is reported for this purpose. Unlike stereotypical assembly either enzymatically polymerized ultralong nucleic acids Mg PPi nanosheets at low ratios or commercial homo‐oligonucleotides devoid biofunctionalities presence metal ions, study represents first demonstration on coordination‐driven high‐efficiency co‐assembly biofunctional hetero‐oligonucleotides non‐Mg nanosheets, specifically glucagon receptor (GCGR) aptamer Co nanosheets. To improve bioavailability nanocomposites, dissolvable microneedle patch applied their delivery. Thereafter, nanocomposites dissociate sustained release 2+ ions GCGR aptamer, from which eliminate colonized bacteria promote angiogenesis through stabilizing HIF‐1α, meanwhile, after circulating to liver reduces blood glucose levels by binding hepatocyte GCGR. Importantly, hypoglycemic effect promotes antibacterial pro‐angiogenic efficacy alleviates inflammatory responses.
