Skin and soft-tissue infections require significant consideration because of their prolonged treatment duration propensity to rapidly progress, resulting in severe complications. The primary challenge stems from the involvement drug-resistant microorganisms that can form impermeable biofilms, as well possibility infection extending deep into tissues, thereby complicating drug delivery. Dissolving microneedle patches are an innovative transdermal drug-delivery system effectively enhances penetration through stratum corneum barrier, increasing concentration at site infection. They offer highly efficient, safe, patient-friendly alternatives conventional topical formulations. This comprehensive review focuses on recent advances emerging trends dissolving-microneedle technology for antimicrobial skin-infection therapy. Conventional antibiotic microneedles compared with those based agents, such quorum-sensing inhibitors, peptides, antimicrobial-matrix materials. also highlights potential incorporating chemodynamic, nanoenzyme antimicrobial, photodynamic, photothermal antibacterial therapies. explores advantages various therapies emphasizes combined application improve efficacy microneedles. Finally, this analyzes druggability different discusses possible future developments.

Abstract The skin microenvironment is a highly intricate and dynamic system, characterized by an acidic pH, diverse microbiota, various metabolites, numerous enzymes, creating both challenges opportunities for the development of innovative drug delivery systems. Dissolving Microneedles (MNs) have emerged as promising, pain‐free alternative to conventional invasive injections, offering ability deliver therapeutics through gradual degradation within skin's interstitial fluids. Building upon unique properties dissolving MNs, novel concept introduced wherein MNs serve in situ chemical reaction chambers. In this framework, can reactants or catalysts skin, enabling initiation specific reactions, such prodrug activation targeted therapy, harmful enhanced synthesis beneficial molecules. Moreover, review systematically explores potential chambers, discussing key aspects their sustained release mechanisms, design strategies, range therapeutic applications. Finally, forward‐looking perspective provided on future addressing broader clinical translation application personalized medicine.

Probiotics such as Lactobacillus and Bifidobacterium spp. have been shown to be critical for maintaining host homeostasis. In recent years, key compounds of postbiotics derived from probiotic metabolism cellular secretion identified their role in organ immunity regulating intestinal inflammation. particular, probiotic-derived extracellular vesicles (PEVs) can act postbiotics, almost the same functional activity probiotics. They also strong biocompatibility loading capacity carry exogenous or parental active molecules reach distal organs play roles. This provides a new direction understanding intrinsic microbiota-host communication mechanism. However, most current studies on PEVs are limited effects/benefits, specific physicochemical properties, composition, mechanisms homeostasis, possible threats remain explored. Here, we review summarize unique properties bioactivities mediating communication, elucidate limitations research potential application postbiotics.
