The primary purpose of this work was to design and obtain a series curcuminoid chalcone-NSAID hybrid derivatives. ester-type compounds with ibuprofen (i), ketoprofen (ii), naproxen (iii) were obtained in two ways, using the Claisen-Schmidt reaction Steglich esterification reaction. designed molecules successfully synthesised, FT-IR, MS, NMR spectroscopy confirmed their structures. Moreover, cytotoxic effect sonodynamic therapy anti-inflammatory, antioxidant, anticholinergic properties some chalcones hybrids evaluated. chalcone derivatives showed promising neuroprotective activity as sonosensitisers for studied cell lines. Additionally, stability determined. RP-HPLC method used observe degradation tested compounds. Studies have shown that structural isomers (3ai, 3bi) are characterised by similar susceptibility factors, i.e., they extremely unstable alkaline environments, very acidic neutral practically stable oxidising photolabile solutions solid phase. These maintain adequate environment at pH 1.2 6.8, which may make them good candidates developing formulations oral administration.

Glioblastoma multiforme (GBM) is one of the most aggressive forms brain tumor, characterized by a daunting prognosis with life expectancy hovering around 12–16 months. Despite century relentless research, only select few drugs have received approval for tumor treatment, largely due to formidable barrier posed blood–brain barrier. The current standard care involves multifaceted approach combining surgery, irradiation, and chemotherapy. However, recurrence often occurs within months despite these interventions. challenges drug delivery overcoming therapeutic resistance become focal points in treatment tumors are deemed essential recurrence. In recent years, promising wave advanced treatments has emerged, offering glimpse hope overcome limitations existing therapies. This review aims highlight cutting-edge technologies ongoing stages development, providing patients valuable insights guide their choices treatment.

Purpose of review Brain tumor treatment presents challenges for patients and clinicians, with prognosis many the most common brain tumors being poor. Focused ultrasound (FUS) can be deployed in several ways to circumvent these challenges, including need penetrate blood–brain barrier spare healthy tissue. This article reviews current FUS applications within neuro-oncology, emphasizing ongoing or recently completed clinical trials. Recent findings Most interest neuro-oncology remains focused on exploring BBB disruption enhance delivery standard-of-care therapeutics. More recently, application radiosensitization, liquid biopsy, sonodynamic therapy is garnering increased attention assist ablation, early detection, phenotypic diagnosis. Preclinical studies show encouraging data immunomodulatory effects FUS, but have yet tested clinically. Summary a burgeoning area research. Data from forthcoming large trials should help clarify its role care.
