Abstract Toxic exposures to heavy metals, such as iron (Fe) and manganese (Mn), can result in long-range neurological diseases are therefore of significant environmental medical concerns. We have previously reported that damage neuroblastoma-derived dopaminergic cells (SH-SY5Y) by both Fe Mn could be prevented pre-treatment with nicotine. Moreover, butyrate, a short chain fatty acid (SCFA) provided protection against salsolinol, selective toxin, the same cell line. Here, we broadened investigation determine whether butyrate might also protect and/or Mn, whether, if combined nicotine, an additive or synergistic effect observed. Both nicotine concentration-dependently blocked toxicities. The ineffective concentrations when combined, full Mn. effects but not mecamylamine, non-selective nicotinic antagonist. Furthermore, potential utility combination metal toxicities is suggested.

Parkinson’s disease (PD) is a progressive neurodegenerative characterized by resting tremor, bradykinesia, rigidity, and postural instability that also includes non-motor symptoms such as mood dysregulation. Dopamine (DA) the primary neurotransmitter involved in this disease, but cholinergic imbalance has been implicated. Current intervention PD focused on replenishing central DA, which provides remarkable temporary symptomatic relief does not address neuronal loss progression of disease. It well established nicotinic receptors (nAChRs) can regulate DA release nicotine itself may have neuroprotective effects. Recent studies identified nAChRs nonneuronal cell types, including glial cells, where they inflammatory responses. Given crucial role neuroinflammation dopaminergic degeneration involvement microglia astrocytes response, provide novel therapeutic target prevention and/or treatment PD. In review, following brief discussion PD, we focus cells and, specifically, their pathology treatment.

Simultaneous enantiomeric analysis is especially important for medicine, food security, and life science. Chiral of multicomponent amine mixtures still faces many challenges. Here, our work demonstrates the first time that a novel chiral derivatizing agent CDApro based on trans-4-fluoro-l-proline (trans4Fpro) has been successfully used rapid simultaneous 22 nonamino acid (non-AA) amines, l/d-AAs, or mirror-image dipeptides in mixture, as well amines with centers several carbons remote to amino group. Furthermore, determination purity quantification can be made using CDApro, which serves robust powerful reagent differentiation amines.

The present study is intended investigate detailed mechanisms related potential neuroprotective effects Nic vivo. Methods: We established a PD model using 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced C57BL6 mice (25 mg/kg/d, 5 d, i.p.) neuropharmacological modulation pretreatment (2.5 i.p., 30 min before MPTP injection) from perspectives neurobehavioral assessment, pathological alterations, microglial cell inflammation and MAPK signaling pathways specific brain regions. Results: open field test, elevated plus maze, rotarod traction test suggested that could significantly improve MPTP-induced motor impairment had an anxiolytic effect. was found neuroapoptosis, enhance tyrosine hydroxylase activity, accumulation phosphorylated α-synuclein substantia nigra striatal regions by TUNEL immunohistochemical assays. Abstract The positive effect of levodopa in the treatment Parkinson’s disease, although it is limited time and has severe side effects, encouraged scientific community to look for new drugs that can stop neurodegenerative process or even regenerate neuromelanin-containing dopaminergic nigrostriatal neurons. L-dopa is a precursor of dopamine used as the most effective symptomatic drug treatment for Parkinson’s disease. Most isolated either synthesized chemically or from natural sources, but only some plants belonging to Fabaceae family contain significant amounts L-dopa. Due its low stability, unambiguous determination in plant matrices requires appropriate technologies. Several analytical methods have been developed different plants. The quantification are mainly based on capillary electrophoresis chromatographic methods, i.e., high-performance liquid chromatography (HPLC), coupled ultraviolet-visible mass spectrometric detection.
