Abstract High-quality point-of-care is critical for timely decision of disease diagnosis and healthcare management. In this regard, biosensors have revolutionized the field rapid testing screening, however, are confounded by several technical challenges including material cost, half-life, stability, site-specific targeting, analytes specificity, detection sensitivity that affect overall diagnostic potential therapeutic profile. Despite their advances in testing, very few classical proven effective commercially viable situations emergency recent COVID-19 pandemic. To overcome these functionalized magnetic nanoparticles (MNPs) emerged as key players advancing biomedical sector with promising applications during ongoing crises. This review focus on understanding developments theranostic (MNPs). Given profound global economic health burden, we discuss impact MNPs acute chronic diseases like small RNA therapeutics, vascular diseases, neurological disorders, cancer, well testing. Lastly, culminate a futuristic perspective scope provide an insight into emerging opportunities whose anticipated to disrupt industry. Graphical

Rationale: Atherosclerosis is characterized by lipid accumulation, plaque formation, and artery stenosis. The pharmacological treatment a promising therapy for atherosclerosis, but this approach faces major challenges such as targeted drug delivery, controlled release, non-specific clearance. Methods: Based on the finding that cathepsin k (CTSK) enzyme enriched in atherosclerotic lesions, we constructed an integrin αvβ3 CTSK-responsive nanoparticle to control release of rapamycin (RAP) locally. The responsive nanoparticles (T/R NPs) were engineered self-assembly targeting polymer PLGA-PEG-c(RGDfC) CTSK-sensitive PLGA-Pep-PEG.PLGA-Pep-PEG was also modified with pair FRET probe monitor hydrolysis events. Results: Our results indicated RAP@T/R NPs accelerated RAP response CTSK stimulation vitro, which significantly inhibited phagocytosis OxLDL cytokines inflammatory macrophages.Additionally, T/R had prolonged blood retention time increased accumulation early late stage atherosclerosis lesions.RAP@T/R blocked development suppressed systemic local inflammation ApoE -/-mice.Conclusions: hold great promise delivery system safer more efficient atherosclerosis.

The inflammatory microenvironment involves changes in pH and protein phosphorylation state is closely related to the occurrence development of atherosclerosis (AS). Herein, we constructed a dual-detection fluorescence nanosensor PCN-NP-HPZ based on post modification MOFs, which realized simultaneous detection imaging through pH-sensitive group piperazine ZrIV node MOFs. sensors were used monitor blood phosphate levels at different time stages during atherosclerotic plaque formation. Two-photon was also performed vascular endothelium. Blood tests combined with two-photon images indicated that early stage AS, tissue lower than normal mice, while higher mice. present study provides new analysis method for assessment disease.

Atherosclerosis, as a life-threatening cardiovascular disease with chronic inflammation and abnormal lipid enrichment, is often difficult to treat timely due the lack of obvious symptoms. In this work, theranostic nanoplatform constructed for noninvasive in vivo diagnosis, plaque-formation inhibition, lesion reversal atherosclerosis. A three-in-one therapeutic complex packaged along polymeric photoacoustic probe into nanoparticles named PLCDP@PMH, which indicates an atherosclerosis-targeting accumulation reactive oxygen species (ROS)/matrix metalloproteinase (MMP) dual-responsive degradation. The suggests lesion-specific imaging on atherosclerotic mice accurate distinct recognition plaques. At same time, performs integrated management through inhibition macrophages M1-polarization, liver X receptor (LXR)-mediated up-regulation ATP-binding cassette transporter A1/G1 (ABCA1/G1) cyclodextrin-assisted dissolution, lead reduced uptake, enhanced efflux, actuated removal. evaluations reveal that PLCDP@PMH can suppress progression further reverse formed plaques under diet without high fat. Hence, provides candidate theranostics early-stage atherosclerosis delivers impressive potential lesions.
