Rationale: Atherosclerosis is characterized by lipid accumulation, plaque formation, and artery stenosis.The pharmacological treatment a promising therapy for atherosclerosis, but this approach faces major challenges such as targeted drug delivery, controlled release, non-specific clearance.Methods: Based on the finding that cathepsin k (CTSK) enzyme enriched in atherosclerotic lesions, we constructed an integrin αvβ3 CTSK-responsive nanoparticle to control release of rapamycin (RAP) locally.The responsive nanoparticles (T/R NPs) were engineered self-assembly targeting polymer PLGA-PEG-c(RGDfC) CTSK-sensitive PLGA-Pep-PEG.PLGA-Pep-PEG was also modified with pair FRET probe monitor hydrolysis events.Results: Our results indicated RAP@T/R NPs accelerated RAP response CTSK stimulation vitro, which significantly inhibited phagocytosis OxLDL cytokines inflammatory macrophages.Additionally, T/R had prolonged blood retention time increased accumulation early late stage atherosclerosis lesions.RAP@T/R blocked development suppressed systemic local inflammation ApoE -/-mice.Conclusions: hold great promise delivery system safer more efficient atherosclerosis.

Atherosclerosis, as a life-threatening cardiovascular disease with chronic inflammation and abnormal lipid enrichment, is often difficult to treat timely due the lack of obvious symptoms. In this work, theranostic nanoplatform constructed for noninvasive in vivo diagnosis, plaque-formation inhibition, lesion reversal atherosclerosis. A three-in-one therapeutic complex packaged along polymeric photoacoustic probe into nanoparticles named PLCDP@PMH, which indicates an atherosclerosis-targeting accumulation reactive oxygen species (ROS)/matrix metalloproteinase (MMP) dual-responsive degradation. The suggests lesion-specific imaging on atherosclerotic mice accurate distinct recognition plaques. At same time, performs integrated management through inhibition macrophages M1-polarization, liver X receptor (LXR)-mediated up-regulation ATP-binding cassette transporter A1/G1 (ABCA1/G1) cyclodextrin-assisted dissolution, lead reduced uptake, enhanced efflux, actuated removal. evaluations reveal that PLCDP@PMH can suppress progression further reverse formed plaques under diet without high fat.
