For the last 40 years, praziquantel has been standard treatment for schistosomiasis, a neglected parasitic disease affecting more than 250 million people worldwide. However, there is no suitable paediatric formulation on market, leading to off-label use and splitting of commercial tablets adults. In this study, we recently available technology, direct powder extrusion (DPE) three-dimensional printing (3DP), prepare Printlets™ (3D printed tablets) amorphous solid dispersions with Kollidon® VA 64 surfactants (Span™ 20 or Kolliphor® SLS). Printlets were successfully from both pellets powders obtained extrudates by hot melt (HME). vitro dissolution studies showed greater four-fold increase in release, due formation dispersions. palatability data indicated that printlets range tolerability, highlighting taste masking capabilities technology without need additional excipients. This work demonstrated possibility 3D using forms HME, avoiding filaments fused deposition modelling 3DP. Moreover, main hurdles praziquantel, such as low drug solubility, inadequate taste, high variable dose requirements, can be overcome technology.

Colonic drug delivery can facilitate access to unique therapeutic targets and has the potential enhance bioavailability whilst reducing off-target effects. Delivering drugs colon requires considered formulation development, as both oral rectal dosage forms encounter challenges if colon's distinct physiological environment is not appreciated. As opportunities surrounding colonic multiply, success of novel pharmaceuticals lies in their design. This review provides a modern insight into key parameters determining effective design development colon-targeted medicines. Influential features governing release, dissolution, stability, absorption are first discussed, followed by an overview most reliable strategies. Finally, appropriate vitro, vivo, silico preclinical investigations presented, with goal inspiring strategic new therapeutics.

3D printing, or additive manufacturing, has gained considerable interest due to its versatility regarding design as well in the large choice of materials. It is a powerful tool field personalized pharmaceutical treatment, particularly crucial for pediatric and geriatric patients. Polysaccharides are abundant inexpensive natural polymers, that already widely used food industry excipients cosmetic formulations. Due their intrinsic properties, such biocompatibility, biodegradability, non-immunogenicity, etc., polysaccharides largely investigated matrices drug delivery. Although an increasing number interesting reviews on manufacturing delivery being published, there gap concerning printing polysaccharides. In this article, we will review recent advances focused applications. Among family polysaccharides, present focus cellulose derivatives, chitosan sodium alginate, printed by fused deposition modeling extrusion-based printing.
