Maternal obesity during pregnancy is associated with adverse outcomes. This might be due to undesired obesity-induced changes in the maternal gut microbiota and related immune adaptations pregnancy. The current study examines how affects immunity pregnant obese lean mice mid-pregnancy (gestational day 12 (GD12)). C57BL/6 were fed a high-fat diet or low-fat from 8 weeks before mating At GD12, we analyzed composition feces responses intestine (Peyer’s patches, mesenteric lymph nodes) peripheral circulation (spleen blood). reduced beneficial bacteria (e.g., Bifidobacterium Akkermansia) changed intestinal dendritic cells, Th1/Th2/Th17/Treg axis, monocytes). Numerous correlations found between obesity-associated bacterial genera intestinal/peripheral anomalies. shows that impacts abundance of specific as compared deranges subsequently change mid-pregnancy. Our findings underscore opportunities for early intervention strategies targeting obesity, ideally starting periconceptional period, mitigate these obesity-related effects.

Disorders of folic acid metabolism during pregnancy lead to fetal neural tube defects (NTDs). However, the mechanisms still require further investigation. Here, we aim analyze brain metabolic profiles 30 NTDs and healthy fetuses. Our results indicated that low-folate diet early life played a causal role in cerebral metabolism, especially lipometabolic disturbance, highlighting importance folate modulating development metabolism. Next, established mouse model NTDs. Interestingly, differential metabolites are mainly involved glycerophospholipid biosynthesis unsaturated fatty acids both human mice brain. Since intestinal microbes could critically regulate neurofunction via intestinal-brain axis, found abundances Firmicutes Bacteroidetes gut pregnant were correlated with lipid related This finding probably reflects intergenerational microbial-metabolism biomarkers
