Sarcopenia, the age-associated decline in skeletal muscle mass and strength, is a condition with complex pathophysiology. Among factors underlying development of sarcopenia are progressive demise motor neurons, transition from fast to slow myosin isoform (type II type I fiber switch), decrease satellite cell number function. Mitochondrial dysfunction has been indicated as key contributor myocyte loss physical performance aging. Several systems have implicated regulation plasticity trophism such fine-tuned between stimulator protein synthesis, mechanistic target rapamycin (mTOR), inhibitor mTOR, AMP-activated kinase (AMPK), that promotes catabolism. Here, we provide an overview molecular mechanisms linking mitochondrial signaling quality homeostasis discuss main pathways elicited by their imbalance during age-related wasting. We also lifestyle interventions (i.e., exercise nutrition) may be exploited preserve function aged muscle. Finally, illustrate emerging possibility rescuing tissue through transplantation.

The associations of dynapenic abdominal obesity and transitions with frailty progression remain unclear among middle-aged older adults. We included 6937 participants from the China Health Retirement Longitudinal Study (CHARLS) 3735 English Aging (ELSA). Participants were divided into non-dynapenia non-abdominal (ND/NAO), alone (AO), dynapenia (D), (D/AO). Frailty status was assessed by index (FI), a linear mixed-effect model employed to analyze D, AO, D/AO, progression. D/AO had increased FI compared ND/NAO in both cohorts. possessed greatest additional annual increase 0.383 (95% CI: 0.152 0.614), followed D AO CHARLS. ELSA magnitude accelerated who transitioned presented CHARLS ELSA. In conclusion, obesity, especially for Our findings highlighted essential intervention targets prevention

Background Research based on observation has demonstrated a relationship between sleep traits and frailty; however, it remains uncertain if this correlation indicates causation. The purpose of study was to look at the causal that exists frailty traits. Method Using summaries from genome-wide association self-reported features index, we performed bidirectional Mendelian randomization (MR) analysis. Examining relationships seven sleep-related goal. major method used calculate effect estimates inverse-variance weighted method, supplemented by median MR-Egger approaches. investigated pleiotropy heterogeneity using several methodologies, such as intercept, MR-PRESSO approach, Cochran’s Q test. We took multivariate genetic correlations related enhance confidence results. Furthermore, MRlap correct for any estimation bias due sample overlap. Results Insomnia, napping during day, apnea syndrome exhibited positive connection with index in forward MR Conversely, there is negative link getting up morning, snoring duration index. During reverse analysis, insomnia, syndrome, while demonstrating duration. There no direct snoring, chronotype, frailty. In MVMR analyses, characteristics indices remained consistent after adjusting potential confounders including BMI, smoking, triglycerides. Conclusion findings our investigation yield novel evidence substantiates notion Through optimization sleep, potentially feasible hinder, postpone, or even state frailty, proposed relevant interventions.

Background: Physical weakness and cardiovascular risk increase significantly with age, but the underlying biological mechanisms remain largely unknown. This study aims to reveal causal effect of circulating metabolites on frailty, sarcopenia vascular aging related traits diseases through a two-sample Mendelian Randomization (MR) analysis. Methods: Exposures were 486 analyzed in genome-wide association (GWAS), while outcomes included sarcopenia, arterial stiffness, atherosclerosis, peripheral disease (PAD) aortic aneurysm. Primary estimates calculated using inverse-variance weighted (IVW) method. Methods including MR Egger, median, Q-test, leave-one-out analysis used for sensitive Results: A total 125 suggestive causative associations between identified. Seven strong links ultimately identified six (kynurenine, pentadecanoate (15:0), 1-arachidonoylglycerophosphocholine, androsterone sulfate, glycine mannose) three (sarcopenia, PAD atherosclerosis). Besides, metabolic pathway 13 significant pathways 6 age-related diseases. Furthermore, metabolite-gene interaction networks constructed. Conclusion: Our research suggested new evidence relationship diseases, which may hold promise as valuable biomarkers.

Background With the rapid growth of global aging, frailty has become a serious public health burden, affecting life quality older adults. Depressive symptoms (depression hereafter) and sleep are associated with frailty, but pathways in which depression affect remain unclear. Method This cross-sectional study included 1866 community-dwelling Demographic characteristics health-related data them was collected, we also assessed depression, quality. Descriptive statistics were carried out ordinal logistic regression analysis used to identify factors correlated frailty. Spearman correlation mediation employed assess associations between quality, Two-sided p < 0.05 considered as significant. Results The results showed that 4.1% adults frail 31.0% pre-frail. Ordinal age, consumptions vegetables, exercise, number chronic diseases, pain, self-rated revealed There effect significant positive predictor (total = 0.0545, 95% boot CI 0.0449–0.0641), mediator (mediation 60.4%). Conclusion Depression poor may be early indicators Improving psychological state can improve is beneficial for healthy aging.

Background: Frailty is a globally recognized issue. However, there lack of evidence exploring factors associated with frailty among older residents in community and nursing-home settings. Methods: To explore the prevalence adults settings, we conducted systematic search following PRISMA guidelines across Web Science, MEDLINE, EMBASE, PubMed, Cochrane databases up until January 2024, selecting 38 studies which encompassed 150,642 participants. Results: Our findings showed higher nursing homes compared to communities. was significantly sociodemographic (living alone, poor self-reported health), physiological (poor sleep, low activity daily living), behavioral (physical inactivity) disease (chronic conditions, depression) both Conclusions: There are numerous These underscore significance promptly identifying high-risk individuals devising appropriate interventions mitigate them.
