Aging is the greatest risk factor for chronic human diseases, including many eye diseases. Geroscience aims to understand effects of aging process on these genetic, molecular, and cellular mechanisms that underlie increased disease over lifetime. Understanding increases general knowledge physiology impacted by processes at various biological extremes. Two major age-related cataract macular degeneration (AMD) are caused dysfunction lens retina, respectively. Lens transparency light refraction mediated fiber cells lacking nuclei other organelles, which provides a unique opportunity study single hallmark, i.e., loss proteostasis, within an environment limited metabolism. In AMD, local photoreceptors/retinal pigmented epithelium/Bruch's membrane/choriocapillaris complex in macula leads photoreceptors eventually central vision, driven nearly all hallmarks shares features with Alzheimer's disease, Parkinson's cardiovascular diabetes. Exosomes are extracellular vesicles ranging from 30 to 150 nm in diameter that contain proteins, nucleic acids and other molecules. Produced by virtually all cell types, they travel throughout the body until reach their target, where can trigger a wide variety of effects transferring molecular cargo recipient cells. In context ocular physiology, exosomes play very important role embryological development, regulation homeostasis immune system, which is crucial for normal vision. Consequently, pathological situations, also undergo modifications terms quantity, composition content, depending on etiology disease. However, mechanisms contribute pathology has not yet been studied depth, many questions remain unanswered. This review aims summarize most recent knowledge function system healthy individuals during processes degenerative, infectious, neurodegenerative, vascular inflammatory nature, such as keratoconus, keratitis, glaucoma, diabetic retinopathy uveitis. Furthermore, given unique characteristics, potential diagnostic biomarkers or therapeutic agents application clinical ophthalmology explored, along with main limitations researchers face today field.

The retinal pigment epithelium (RPE) is a highly specialised monolayer subjected to constant oxidative stress, which, in the long term, favours development of complex pathological process that underlying cause macular damage. Therefore, counteracting overproduction ROS best-researched approach preserve functional integrity RPE. S-Petasin, secondary metabolite extracted from plant Petasites hybridus, has numerous biological effects, which highlight its anti-inflammatory and antioxidative properties. aim our study investigate whether S-Petasin exerts cytoprotective effects by protecting RPE pretreatment with were assessed determination cell viability, intracellular levels, activation Nrf2 pathway resulting post-transcriptional antioxidant/antiapoptotic response. Our results show (1) reduces improving viability exposed damage; (2) activates signalling pathway, modulating response antioxidant chemical biomarkers; (3) Bax an increase those Bcl-2, concomitant downregulation Bax/Bc-2 ratio. This study aimed to explore the relationship between biochemical profiles and risk of age-related macular degeneration (AMD) through a cross-sectional observational analysis. We examined data U. S. population from 2005–2008 National Health Nutrition Examination Survey (NHANES) database. Student's t-test, multivariable logistic regression, Pearson's correlation, restricted cubic spline (RCS) model, linear regression were applied analyze underlying AMD risk, comparing non-AMD subgroups. Multivariable adjusted for age demographic factors, showed no significant associations levets specific parameters (P > 0.05). correlation revealed positive total bilirubin, uric acid in subgroup < 0.05), but such liner association was found The RCS model confirmed non-linear relationships presented these variables subgroup. In addition, without adjustment, acid, presence Biochemical profiles, after adjusting age, did not significantly influence risk. However, bilirubin might potentially be related presence. Our findings suggest need further research clarify role biomarkers development.

Immunosenescence, the slow degradation of immune function over time that is a hallmark and driver aging, makes older people much more likely to be killed by common infections (such as flu) than young adults, but it also contributes greatly rates chronic inflammation in later life. Such micro nutrients are crucial for modulating effective responses their deficiencies have been associated with dysfunctional immunity elderly. In this review, we specifically focused on contribution major (Vitamins A, D E, Vitamin C; Zinc Selenium) immunomodulators ageing population especially related inflame-ageing process including autoimmunity. This review will cover these hologenomic interactions, how can modulate cell and/or cytokine production benefit hosts healthy mucous-associated along sustainable immunologic homeostasis. For example, points out modulatory effects vitamin both innate adaptive immunity, specific focus its ability suppress pro-inflammatory cytokines synthesis while enhancing regulatory T-cell function. same context, zinc described important nutrient thymic differentiation exhibits immunomodulatory functions decreasing inflammation. addition, go increase systemic low-grade and, inflammaging well actually enhance autoimmune pathologies old age. It assesses potential role additional targeted nutritional supplementation counteract effects, promoting wider resilience adults. collates current evidence highlights adequate intake autoimmunity during ageing, providing plausible origins interventions promote aging.
