Ulcerative colitis (UC) is a complex immune-mediated chronic inflammatory bowel disease. It mainly characterized by diffuse inflammation of the colonic and rectal mucosa with barrier function impairment. Identifying new biomarkers for development more effective UC therapies remains pressing task current research. Ferroptosis newly identified form regulated cell death iron-dependent lipid peroxidation. As research deepens, ferroptosis has been demonstrated to be involved in pathological processes numerous diseases. A growing body evidence suggests that pathogenesis associated ferroptosis, regulation provides opportunities treatment. However, specific mechanisms which participates remain fully thoroughly investigated. Therefore, this review, we focus on advances mechanism recent years describe potential role UC. In addition, explore underlying crosslinked pathway between other such as macrophages, neutrophils, autophagy, endoplasmic reticulum stress, gut microbiota Finally, also summarize compounds may act inhibitors future.

Abstract The gut microbiota plays a critical role in maintaining human health, influencing wide range of physiological processes, including immune regulation, metabolism, and neurological function. Recent studies have shown that imbalances composition can contribute to the onset progression various diseases, such as metabolic disorders (e.g., obesity diabetes) neurodegenerative conditions Alzheimer's Parkinson's). These are often accompanied by chronic inflammation dysregulated responses, which closely linked specific forms cell death, pyroptosis ferroptosis. Pathogenic bacteria trigger these death pathways through toxin release, while probiotics been found mitigate effects modulating responses. Despite insights, precise mechanisms influences diseases remain insufficiently understood. This review consolidates recent findings on impact immune‐mediated inflammation‐associated conditions. It also identifies gaps current research explores potential advanced technologies, organ‐on‐chip models microbiome–gut–organ axis, for deepening our understanding. Emerging tools, single‐bacterium omics spatial metabolomics, discussed their promise elucidating microbiota's disease development.

Background: Ulcerative colitis (UC) is a chronic intestinal disease of growing global concern. Bacteria associated with fermented food or probiotics regulate immune and inflammatory responses, playing key role in homeostasis. Results: Five relatively good antioxidant effects, namely Lactiplantibacillus plantarum H6, Latilactobacillus sakei QC9, Limosilactobacillus fermentum E7, Bacillus subtills D1, licheniformis Q13, were screened out from 30 strains through vitro assays. The five had varying degrees alleviating effects on UC mice improved various physiological indicators, such as oxidative stress parameters histopathological sections. Q13 more pronounced. Furthermore, E7 effectively regulated mouse microbiota composition, increased short-chain fatty acid concentration, promoted the expression anti-inflammatory factors, interleukin 10 (IL-10), while suppressing that pro-inflammatory 1β (IL-1β), 6 (IL-6), tumor necrosis factor α (TNF-α). Meanwhile, D1 only exhibited partial effects. Finally, tight junction proteins colon tissues. Conclusions: showed superior efficacy to other UC, offering novel therapeutic prospects for safer effective management UC.

Abstract Ferroptosis is a novel form of lipid peroxidation-driven, iron-dependent programmed cell death. Various metabolic pathways, including those involved in and iron metabolism, contribute to ferroptosis regulation. The gut microbiota not only supplies nutrients energy the host, but also plays crucial role immune modulation balance. In this review, we explore pathways associated with impact on host metabolism. We subsequently summarize recent studies influence regulation by discuss potential mechanisms through which affects ferroptosis. Additionally, conduct bibliometric analysis relationship between context chronic kidney disease. This can provide new insights into current research status future microbiota.

Large-scale studies indicate a strong relationship between the gut microbiome, type 2 diabetes mellitus (T2DM), and atherosclerotic cardiovascular disease (ASCVD). Here, higher abundance of III secretion system (T3SS) virulence factors Enterobacteriaceae/Escherichia-Shigella in patients with T2DM-related-ASCVD, which correlates their stenosis is reported. Overexpression T3SS via Citrobacter rodentium (CR) infection Apoe-/- T2DM mice exacerbated lesion formation increased permeability. Non-targeted metabolomic proteomic analysis mouse serum showed that caused abnormal glycerophospholipid metabolism mice. Proteomics, RNA sequencing, functional analyses induced ferroptosis intestinal epithelial cells, partly due to expression ferritin heavy chains (FTH1). This findings first demonstrated increases disrupting barrier upregulation phosphatidylcholine, thereby exacerbating T2DM-related ASCVD.
