Background: COVID-19 is a disease in several stages starting with virus replication to dysregulation immune system response, organ failure and recovery/death. Our aim was determine the effect of Ganoderma lucidum, lycopene, sulforaphane, royal jelly resveratrol extract on markers oxidative stress, inflammation, routine laboratory analyses duration symptoms patients. Methods: The stress parameters interleukines 6 8 (IL-6, IL-8), tumor necrosis factor alpha (TNF-α) were determined order estimate antioxidant anti-inflammatory product using spectrophotometric magnetic bead-based multiplex assay serum 30 patients mild form COVID-19. Results: Statistically significant differences obtained for all investigated between treated control group. Moreover, observed leukocytes, neutrophil leukocyte ratio iron. average 9.4±0.487 days versus 13.1±0.483 treatment group, respectively (p=0.0003). Conclusion: results demonstrated promising Ge132+NaturalTM reducing IL-6, IL-8 TNF-α levels, evidence presented herein suggest that combination lucidum extract, could be used as potent an adjuvant therapy diseases accompanied by increased inflammation.

Chronic inflammation is an important component of many diseases, including autoimmune intracellular infections, dysbiosis and degenerative diseases. An element this state the mainly positive feedback between inflammatory cytokines, reactive oxygen species (ROS), nitric oxide (NO), increased calcium, hypoxia-inducible factor 1-alpha (HIF-1α) stabilisation mitochondrial oxidative stress, which, under normal conditions, enhance response against pathogens. Autophagy nuclear erythroid 2-related 2 (Nrf2)-mediated antioxidant are negatively coupled with above-mentioned elements to maintain defence at a level appropriate severity infection. The current review first attempt build multidimensional model cellular self-regulation chronic inflammation. It describes feedbacks involved in explains possible pathways by which becomes chronic. multiplicity suggests that symptomatic treatment should focus on inhibiting multiple effectively suppress all dysregulated inflammation, calcium mito-stress other metabolic disturbances.

The SARS-CoV-2 proteases Mpro and PLpro are critical targets for antiviral drug development the treatment of COVID-19. 1,2,4-thiadiazole functional group is an inhibitor cysteine proteases, such as papain cathepsins. This chemical moiety also present in ceftaroline fosamil (CF), FDA-approved fifth-generation cephalosporin antibiotic. study investigates interactions between CF, its primary metabolites (M1 dephosphorylated CF M2 opened β-lactam ring) derivatives (protonated M1H M2H), open rings (open-M1H open-M2H) with evaluates CF’s effects on vitro viral replication. In silico analyses (molecular docking molecular dynamics (MD) simulations) demonstrated that potential inhibitors Mpro. Docking analysis indicated majority ligands were more stable than PLpro; however, biochemical preferred target CF. inhibited replication human Calu-3 cell model at submicromolar concentrations when added to culture medium 12 h. Our results suggest should be evaluated a repurposing agent COVID-19, considering not only but other relevant cellular pathways. Additionally, reactivity sulfur warrants further exploration protease inhibitors.

Coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2 is characterized acute respiratory distress syndrome (ARDS) facilitated cytokine storm and other risk factors that increase susceptibility complications leading to death. Emerging as a major global public health challenge, the has claimed more than 6 million lives catastrophic economic disruptions. However, there are concerns about safety well efficacy of drugs vaccines presently used control pandemic, therefore necessitating intense search for safe natural products can effectively safely combat it. This work reviews studies on lingzhi or reishi medicinal mushroom, Ganoderma lucidum its properties may potentially infection co-morbidities. Available evidence suggests Ganoderma mushroom co-morbidities aggravate severity disease. Preclinical clinical evaluation establish dose, efficacy, potential toxicity possible use in management COVID-19 recommended.

The global COVID-19 pandemic, caused by SARS-CoV-2, has created an urgent need for novel antiviral agents, as it led to a devastating loss of lives worldwide. In response, numerous vaccines were developed in 2022, with ongoing efforts more candidates. Despite significant progress, reports vaccine-related issues and side effects have emerged. effectiveness been challenged new viral variants, lack well-designed vivo experiments, randomised controlled trials. Currently, no clinically approved medications exist treating COVID-19. However, few broad-spectrum drugs shown promising outcomes clinical studies. Many scientists believe that natural compounds derived from herbal medicines mushrooms could offer substantial benefits patients. Ganoderma, known its traditional use promoting health longevity, rich history therapeutic applications dietary supplementation. Numerous epidemiological studies demonstrated Ganoderma derivatives can enhance innate adaptive immune responses against various pathogens, including viruses, laboratory settings living organisms. Therefore, the secondary metabolites such triterpenoids, polysaccharides, glycoproteins, hold potential developing drugs. These mechanisms, inhibition replication, modulation responses, direct virucidal effects. Given search effective treatments COVID-19, bioactive stand out candidates further research development. Their multifaceted mechanisms make them suitable exploration agents potentially contributing enhancing existing treatments.
