Stingless bee propolis has emerged globally as a new source of bioactive molecules that can advance human health. However, limited research been conducted on Australian stingless propolis. This study investigated the chemical composition and biological activity produced by bees Tetragonula carbonaria from Gibberagee, distinct region New South Wales state in Australia. Using bioassay-guided fractionation, twelve compounds were isolated, including six A-ring methylated flavonoids. Nine these demonstrated strong scavenging against 2,2-diphenyl-1-picrylhydrazyl radicals, with five exhibiting greater potency than vitamin C. Chemical structures seven additional minor flavonoids determined through an intensive MS/MS data analysis. In silico screening 19 revealed all, except for gallic acid, displayed higher binding affinity to α-glucosidase antidiabetic drug, voglibose. showed Gibberagee is promising nutraceutical cosmeceutical applications owing its antioxidant properties. Propolis is a resinous mixture produced by honeybees which has been used since ancient times for its useful properties. Acute pancreatitis (APS) is a prevalent acute pancreatic inflammation, where oxidative stress, inflammatory signaling pathways, and apoptosis activation contribute to injury. Pinocembrin, the predominant flavonoid in propolis, was explored for its likely shielding effect against APS provoked by two intraperitoneal doses of L-arginine (250 mg / 100 g) rat model. Pinocembrin ameliorated histological immunohistochemical changes tissues lowered activities amylase lipase that were markedly elevated with administration. Moreover, pinocembrin reinstated oxidant/antioxidant equilibrium, which perturbed L-arginine, boosted levels nuclear factor erythroid 2-related 2 (Nrf2) heme oxygenase-1 (HO-1). reduced elevation serum C-reactive protein (CRP) level induced L-arginine. Additionally, it decreased expression high motility group box 1 (HMGB1), toll-like receptor 4 (TLR4), kappa B (NF-κB), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), NOD-like (NLR) Family Pyrin Domain Containing 3 (NLRP3) inflammasome pancreas.
