Temperature plays a significant role in anaerobic digestion (AD) as it affects the microbial communities and ultimately controls efficiency of process. Few studies have looked at temperature-adjusted AD, but is unclear how temperature shifts affect biogas production dynamics microorganisms involved methanogenesis. This study tested two shift scenarios fed-batch mode using anaerobically digested sewage sludge glucose-based substrate. The first scenario was acclimatized to upshifting temperatures from 42 °C 48 while second downshifting 55 45 °C. Both resulted decrease production, especially upshifted experienced maximum 83%, downshifted 16–33% methane production. Next-generation 16S rRNA sequencing revealed domination Methanoculleus scenario. However, low correlation between number other hydrogenotrophic methanogens indicates inhibition pathway. showed better due substantial acetoclastic Methanosaeta abundance sulfate-reducing bacteria. Hence, communities, significantly affecting performance.

Compared to the conventional single-stage AD process, two-stage process with separate H2 and CH4 production provides higher energy efficiency enhanced operation stability.
