Fluorescent sensor is an important tool to reliaze qualitative or quantitative detection of target analyte based on the fluorescence principle. Clustered regularly interspaced short palindromic repeats/CRISPR-associated protein (CRISPR/Cas) has been utilized develop as a precise, efficient, and highly sensitive molecular diagnostic due its efficient targeting gene editing ability. At present, CRISPR/Cas system-based fluorescent sensors have shown excellent performance in field analysis detection, received widespread attention. Therefore, this paper reviews mechanism system, characteristics different Cas proteins, principle sensor, with focus summarizing application for detection.

A microfluidic chip is a tiny reactor that can confine and flow specific amount of fluid into channels tens to thousands microns as needed precisely control flow, pressure, temperature, etc. Point-of-care testing (POCT) requires small equipment, has short cycles, controls the process, allowing single or multiple laboratory facilities simultaneously analyze biological samples diagnose infectious diseases. In general, rapid detection stage assessment viral epidemics are essential overcome pandemic situations promptly. Therefore, combining devices with POCT improves efficiency convenience for disease SARS-CoV-2. At same time, chips increases user accessibility, accuracy sensitivity, shortens etc., which beneficial in detecting This review shares recent advances POCT-based COVID-19 how it better suited help response ongoing pandemic.

The CRISPR/Cas system is a powerful genome editing tool and possesses widespread applications in molecular diagnostics, therapeutics genetic engineering. But easy folding of the target sequences causes remarkable deterioration recognition shear efficiency case single Cas-CRISPR RNA (crRNA) duplex. Here, we develop cooperative shearing (CRISPR-CS) system. Compared with traditional system, two CRISPR/Cas-crRNA duplexes simultaneously recognize different sites sequence, increasing possibility efficiency. Cooperative cuts more methylene blue-ssDNA reporters on electrode, enabling unamplified nucleic acid electrochemical assay less than 5 minutes detection limit 9.5×10
