Daphne mezereum L., an important medicinal plant in Scandinavian folk medicine, was used to treat ailments such as diarrhea, swelling and stomach pain. A range of natural compounds have been isolated, but little attention has given the polysaccharides this plant. Previous work our group shown that a polysaccharide enriched fraction from bark D. exhibited pro-inflammatory effects. To pursue further, aim present isolate characterize these polysaccharides. From ethanol-precipitate water extract, one neutral (DMP-NF) acidic (DMP-AF) isolated by anion-exchange chromatography. GC, GC-MS 1D- 2D-NMR were structures. DMP-NF appeared be mixture arabinan, arabinogalactan hemicelluloses xyloglucan, mannan xylan. DMP-AF contained pectic mainly consisting unusually long homogalacturonan backbone. Enzymatic treatment pectinase yielded DMP-ED, which rhamnogalacturonan-I backbone with galactan side chains. Both DMP-ED induced IFN-γ TNF-α secretion peripheral blood mononuclear cells (PBMCs), being most potent fraction. less active, might due sterically available domain.

The use of natural polysaccharides as biomaterials is gaining importance in tissue engineering due to their inherent biocompatibility. In this direction, the present study aims explore structure and biocompatibility EPS produced by Virgibacillus dokdonensis VITP14. This marine bacterium produces 17.3 g/L at 96 h fermentation. was purified using ion exchange gel permeation chromatographic methods. porous web-like elemental composition (C, O, Na, Mg, P, S) were inferred from SEM EDX analysis. AFM analysis revealed spike-like lumps with a surface roughness 84.85 nm. zeta potential value −10 mV indicates anionic nature EPS. Initial molecular characterization showed that heteropolysaccharide composed glucose (25.8%), ribose (18.6%), fructose (31.5%), xylose (24%), which are monosaccharide units HPLC FTIR spectrum presence functional groups/bonds typical EPSs (O-H, C-H, C-O-H, C-O, S=O, P=O). polymer has an average weight 555 kDa. Further, NMR monomer composition, existence two α- six β-glycosidic linkages, branched repeating unit → 1)[α-D-Xylp-(1 2)-α-D-Glcp-(1 6)-β-D-Glcp-(1 5)]-β-D-Frup-(2 2)[β-D-Xylp-(1 4)]-β-D-Xylp-(1 6)-β-D-Fruf-(2 4)-β-D-Ribp-(1 →. thermally stable till 251.4 °C. X-ray diffraction confirmed semicrystalline (54.2%) exhibits significant water solubility (76.5%), water-holding capacity (266.8%), emulsifying index (66.8%), hemocompatibility (erythrocyte protection > 87%), cytocompatibility (cell viability 80% on RAW264.7 keratinocyte HaCaT cells) higher concentrations prolongs coagulation time APTT PT tests.
