Cyclodextrins (CD) and derivatives are functional excipients that can improve the bioavailability of numerous drugs. Because their drug solubility improving properties they used in many pharmaceutical products. Furthermore, stability small molecular drugs be improved by incorporation CDs an unpleasant taste smell masked. In addition to well-established CD including hydroxypropyl-β-CD, hydroxypropyl-γ-CD, methylated- β-CD sulfobutylated- β-CD, there promising new development. particular, CD-based polyrotaxanes exhibiting cellular uptake enhancing properties, CD-polymer conjugates providing sustained release, enhanced uptake, mucoadhesive thiolated showing mucoadhesive, situ gelling, as well permeation will likely result innovative delivery systems. Relevant clinical trials showed various applications such formation nanoparticles, stabilizing for protein or development ready-to-use injection Advanced products making use benefical at same time. Within this review we provide overview on these recent developments take outlook how class further shape landscape delivery.

This study aimed to evaluate the effect of thiolated micelles on cellular uptake their payload. Reduced and oxidized glutathione were covalently attached palmitic acid via amide bond formation. Micelles formed with these surfactants (MP-GSH MP-GSSG-P) evaluated regarding critical micellar concentration (CMC) hemolytic activity. Cytotoxicity was HEK 293 HeLa cells. Diffusion in cells by fluorescence correlation spectroscopy (FCS). Furthermore, containing coumarin-6 as model drug analyzed flow cytometry confocal laser scanning microscopy. CMC MP-GSSG-P, MP-GSH, (MPA) determined be 0.455 mM, 0.166 0.046 respectively. At a 0.5 % MPA caused around 80 hemolysis. In cells, MP-GSSG-P exhibited toxicity at 0.25 %, while MP-GSH showed 0.06 after 4 hours incubation. contrast, more resilient, only showing %. Diffusivity within higher than that MPA. Cellular studies demonstrated significantly (p < 0.05) enhanced internalization 112-fold 270-fold 12-fold 60-fold when compared These findings suggest could serve promising vehicles for enhancing drugs.

Psoriasis is a chronic and recurrent inflammatory disease driven not only by intrinsic factors such as immune system dysregulation but also external factors, including bacterial infections. In contrast to the control of single pathogenic pathway, combination therapies addressing both infectious components psoriasis pathogenesis may offer more effective strategy for controlling its progression. this study, we developed sprayable hydrogel incorporating tea polyphenol-loaded lauric acid liposomes (TP@LA-Lipo gel) investigate anti-inflammatory antibacterial role in psoriasis. Our results demonstrated that TP@LA-Lipo modulated macrophage activity, reduced expression iNOS TNF-α, remodeled microenvironment. Meanwhile, effectively eliminated Staphylococcus aureus Escherichia coli through membrane disruption, mitigating provoked response. More importantly, gel, when sprayed onto lesions, provided sustained drug release over three days, enabling deeper penetration thickened stratum corneum reach inflamed layers beneath. Furthermore, an imiquimod-induced mouse model, gel restored damaged skin, alleviated histopathological changes, systemic summary, these findings indicate offers comprehensive management improving quality life patients.

Overcoming P-glycoprotein (P-gp)-mediated efflux poses a significant challenge for the pharmaceutical industry. This study investigates potential of thiolated β-cyclodextrins (β-CD-SHs) as inhibitors P-gp-mediated in Caco-2 cells. Through series transport assays, intracellular accumulation, and P-gp substrates Rhodamine 123 (Rh123) Calcein-AM with without co-administration β-CD-SHs were assessed. The results revealed that cellular uptake Rh123 enhanced up to 7- 3-fold, compared control, respectively. In studies an 2.5-fold reduction was reached indicating substantial decrease by β-CD-SHs. Furthermore, it observed led reactivity fluorescence-labeled anti-P-gp, suggesting additional effects on conformation P-gp. Overall, this demonstrates effective modulator drug
