Exosomes are extracellular vesicles of endosomal origin that released by practically all cell types across metazoans. active vehicles intercellular communication and can transfer lipids, RNAs, proteins between different cells, tissues, or organs. Here, we describe a mechanism whereby containing KFERQ motif pentapeptide loaded into subpopulation exosomes in process is dependent on the membrane protein LAMP2A. Moreover, demonstrate this independent ESCRT machinery but HSC70, CD63, Alix, Syntenin-1, Rab31, ceramides. We show master regulator hypoxia HIF1A to transport signaling normoxic cells. In addition, tagging fluorescent with KFERQ-like sequences, were able follow interorgan exosomes. Our findings open new avenues for exosome engineering allowing loading bioactive them motifs.

Abstract Liquid biopsy, characterized by minimally invasive detection through biofluids such as blood, saliva, and urine, has emerged a revolutionary strategy for cancer diagnosis prognosis prediction. Exosomes are subset of extracellular vesicles (EVs) that shuttle molecular cargoes from donor cells to recipient play crucial role in mediating intercellular communication. Increasing studies suggest exosomes have great promise serve novel biomarkers liquid since large quantities enriched body fluids involved numerous physiological pathological processes. However, the further clinical application been greatly restrained lack high-quality separation component analysis methods. This review aims provide comprehensive overview on conventional technologies exosome isolation, characterization content detection. Additionally, roles serving potential biopsy diagnosis, treatment monitoring, prediction summarized. Finally, prospects challenges applying exosome-based precision medicine evaluated.

Abstract Extracellular vesicles (EVs) are nano-sized, membranous structures secreted into the extracellular space. They exhibit diverse sizes, contents, and surface markers ubiquitously released from cells under normal pathological conditions. Human serum is a rich source of these EVs, though their isolation proteins non-EV lipid particles poses challenges. These transport various cellular components such as proteins, mRNAs, miRNAs, DNA, lipids across distances, influencing numerous physiological events, including those within tumor microenvironment (TME). Their pivotal roles in communication make EVs promising candidates for therapeutic agents, drug delivery systems, disease biomarkers. Especially cancer diagnostics, EV detection can pave way early identification offers potential diagnostic Moreover, subtypes emerging targeted tools, highlighting clinical significance. The need non-invasive biomarkers to monitor biological processes purposes remains unfulfilled. Tapping unique composition could unlock advanced avenues future. In this review, we discuss detail conditions, cancers (encompassing head neck, lung, gastric, breast, hepatocellular carcinoma), neurodegenerative disorders, diabetes, viral infections, autoimmune renal diseases, emphasizing advancements molecular diagnostics delivery.

Primary lung cancer is one of the most common malignant tumors in China. Approximately 60% patients have distant metastasis at initial diagnosis, so it necessary to find new tumor markers for early diagnosis and individualized treatment. Tumor contribute play important roles detection treatment, as well precision medicine, efficacy monitoring, prognosis prediction. The pathological small biopsy specimens determines whether there are cells type. Because traumatic compliance with multiple biopsies poor, liquid has become a hot research direction. Liquid advantageous because they nontraumatic, easy obtain, reflect overall state tumor, allow real-time monitoring. At present, mainly include circulating cells, DNA, exosomes, microRNA, RNA, platelets, endothelial cells. This review introduces progress clinical application prospect technology cancer.
