Bladder cancer (BC) is one of the most common cancers in world, with high morbidity and mortality. It essential to develop a non-invasive, highly accurate, simple method for BC diagnosis. This work proposed fluorescent biosensor based on inorganic nanoflares combined DNAzyme walker simultaneous detection exosomal microRNAs (miRNAs). was constructed Au nanoparticle (AuNP) modified carbon dot (CD)-labeled substrates strands (AuNP@CDs nanoflares-DNAzyme, APCD). In presence target miRNAs, activated then cleaved CD-labeled automatically walked along AuNP, allowing fluorescence recovery. Due structure functional composition, APCD biosensors demonstrated sensitivity specificity, reached limit single miRNA at femtomolar level wide linear range from 50 fM 10 nM. Furthermore, analysis BC-related miR-133b miR-135b clinical serum specimens achieved consistent qRT-PCR, suggesting it potential diagnosis other cancers.

Extracellular vesicles (EVs) such as ectosomes and exosomes have gained attention promising natural carriers for drug delivery. Exosomes, which range from 30 to 100 nm in diameter, possess a lipid bilayer are secreted by various cells. Due their high biocompatibility, stability, low immunogenicity, favored cargo carriers. The membrane of also offers protection against degradation, making them desirable candidate However, loading into remains be challenge. Despite strategies incubation, electroporation, sonication, extrusion, freeze–thaw cycling, transfection that been developed facilitate loading, inadequate efficiency still persists. This review an overview current delivery using summarizes recent approaches small-molecule, nucleic acid, protein drugs exosomes. Central nervous system (CNS) diseases, such as multiple sclerosis, Alzheimer's disease (AD), and Parkinson's (PD), affect millions of people around the world. Great efforts were put in related research, but few breakthroughs have been made diagnostic therapeutic approaches. Exosomes are cell-derived extracellular vesicles containing diverse biologically active molecules secreted by their cell origin. These contents, including nucleic acids, proteins, lipids, amino metabolites, can be transferred between different cells, tissues, or organs, regulating various intercellular cross-organ communications normal pathogenic processes. Considering that cellular environment state strongly impact content uptake efficiency exosomes, detection biological fluids composition analysis potentially offer a multicomponent readout several human diseases. Recently, studies found aberrant secretion exosomes closely to pathogenesis CNS Besides, loading natural cargoes, deliver drugs cross blood brain barrier, making them emerging candidates biomarkers therapeutics for In this review, we summarize discuss advanced research progress pathological processes diseases regarding with neuroinflammation, repair, protein aggregation.
