Implant-associated infections and excessive immune responses are two major postsurgical issues for successful implantation. However, conventional strategies including antibiotic treatment inflammatory regulation always compromised due to the comodification of various biochemical agents instances functional interference. It is imperative provide implant surfaces with satisfactory antibacterial anti-inflammatory properties. Here, a dual-effect nanostructured polyetheretherketone (PEEK) surface (NP@PDA/Zn) bionic mechano-bactericidal nanopillars immobilized immunomodulatory Zn2+ designed. The constructed hybrid display remarkable performance against Gram-negative Gram-positive strains through synergy physical chemical bactericidal effects imposed by Zn2+. Meanwhile, immunoregulatory property evaluated investigation macrophage polarization both in vitro vivo, results reveal that NP@PDA/Zn could downregulate expression M1-related cytokines decrease M1 recruitment lower response. Notably, exhibited exceptional biocompatibility discerning biocidal activity between bacterial mammalian cells antioxidant effectively scavenges ROS, minimizing potential cytotoxicity. Taken together, presents convenient promising strategy combining synergistic without any mutual interference, which can support development multifunctional implant-associated materials.

Abstract The morphology of regular and uniform arrays black silicon structures was evaluated for bactericidal efficacy against gram-positive, non-motile Staphylococcus epidermidis ( S. ). In this study, were fabricated using nanosphere lithography deep reactive ion etching. effects nanomorphology on bacterial killing systematically nanostructures with pitches ranging from 300 to 1400 nm pitch spherical cocci approximately 500 1000 in diameter. Our results show that nanostructure factors such as height roughness do not directly determine efficacy. Implant-associated infections are significant impediments to successful surgical outcomes, often resulting from persistent bacterial contamination. It has been hypothesized that bacteria can transfer electrons semiconductors with comparable potential the biological redox (BRP). Building on this concept, we developed an antibiotic-free bactericidal system, Co3O4/TiO2-Ti, capable of achieving real-time and sustainable effects. Our study demonstrated possessing appropriately set valence band, initiated charge transfer, reactive oxygen species (ROS) production, membrane damage in adherent Staphylococcus aureus (S. aureus). Notably, vivo experiments illustrated remarkable antibacterial activity while promoting soft-tissue reconstruction demonstrating excellent cytocompatibility. Transcriptomic analysis further revealed a down-regulation aerobic respiration-associated genes up-regulation ROS-associated S. presence Co3O4/TiO2-Ti compared Ti. Kyoto Encyclopedia Genes Genomes (KEGG) gene enrichment (GSEA) identified alterations respiratory metabolism, oxidative phosphorylation, synthesis amino acid cultured Co3O4/TiO2-Ti. Furthermore, when combined near-infrared (NIR) irradiation photothermal therapy (PTT), eliminated 95.71% floating vitro. The findings suggest strategy holds substantial promise enhancing implant sterilization capabilities, thereby contributing prevention treatment through bandgap engineering implants NIR irradiation.
